免疫吸附磁性涂层碳纤维的研究

【摘要】 近年来,免疫吸附治疗技术已成为去除血液中致病物质的一种重要手段。本项目把磁性微球固定在有良好的生物相容性的碳纤维表面,制备出免疫吸附磁性涂层碳纤维载体。主要结论如下:(1)制备有机溶胶过程中,产物为较高纯度的纳米Fe3O4且Fe3O4颗粒外层包覆了油酸。相对最佳的制备路线为在反应阶段同时滴加表面改性剂。油酸用量在0.7ml时,既能充分包覆Fe3O4颗粒,又能保持较好的稳定性、分散性。制备水基溶胶过程中,采用不同表面活性剂得到的产物都为纯度较高的Fe3O4,且改性后的Fe3O4颗粒表面都包覆了相应的表面改性剂。聚乙二醇作为表面改性剂制备的水基Fe3O4溶胶的分散性和稳定性相对最好。(2)有机基Fe3O4溶胶制备的磁性涂层碳纤维由碳和Fe3O4两种物相组成,且表面光滑,涂层均匀。而在各水基溶胶中,聚乙二醇作为表面改性剂制备的Fe3O4微球在碳纤维表面涂覆效果最好。(3)静态平行吸附试验表明,进行了表面磁性处理和活化偶联的碳纤维对BSA(牛白蛋白)的吸附远高于空白活性碳纤维,且吸附性能相对稳定。并且聚乙二醇水基Fe3O4溶胶制备的免疫吸附磁性涂层碳纤维的吸附量相对高于有机基溶胶制备的。通过正交试验,得到了活化偶联过程中各因素对吸附量的影响的主次顺序,获得相对最好的水平搭配。(4)在设计的模拟免疫吸附柱试验中,两种装填方式并不理想而淘汰,由此提出了另一种设计,将制备好的免疫吸附磁性涂层碳纤维编织成过滤布。

【Abstract】 In recent years, immunoadsorption therapy has become an important Method to Removal of pathogenic substances in blood. In this project, magnetic microspheres were fixed on the surface of the good biocompatibility carbon fibers to prepare immunoadsorption magnetic coated carbon fiber carrier.The main conclusions are as follows:(1) In process of preparation organic sol, the product is high purity nano-Fe3O4 and the outer layers of Fe3O4 particles are coated with oleic acid. The best preparation route is dropping the surface modifier during the synthesis reaction. The Fe3O4 particles can be fully coated by oleic acid to maintain good stability and dispersion when the oleic acid dosage is 0.7ml. In process of preparation Hydrosol, the products using different surfactants are high purity Fe3O4, and the modified surface of Fe3O4 particles are coated with a corresponding surface modifier. The dispersion and stability of the Fe3O4 hydrosol prepared by PEG as surface modifier was the best.(2) The magnetic coated carbon fiber prepared by Fe3O4 organic sol is composed of carbon and Fe3O4, and has smooth surface and uniform coating. In various Fe3O4 hydrosols, the Fe3O4 microspheres prepared by PEG as a surface modifier have the best coating on the surface of carbon fiber.(3) The static parallel adsorption test shows that the adsorption on the BSA of activated carbon fibers with magnetic surface treatment and activating-coupling treatment is much higher than the blank activated carbon fiber and the adsorption property is relative stability. The adsorption of immunoadsorption magnetic coated carbon fiber prepared by Fe3O4 hydrosol that PEG as surface modifier is relatively higher than prepared by Fe3O4 organic sol preparation. By orthogonal test, there are obtained the important order of the factors on the adsorption property in activation-coupling process, and gotten the best level combination.(4) In the simulation experiment of immunosorbent column, two kinds of packing structure were eliminated because of unsatisfactory, thus an alternative design was proposed, which is knitting immunoadsorption magnetic coated carbon fibers into filter cloth.
