双峰驼自然发酵乳对小鼠抗应激和抗氧化能力的影响

【摘要】 为探讨双峰驼自然发酵乳(酸驼乳)的抗应激和抗氧化作用,本实验以清洁级昆明种小白鼠为实验动物,给予不同剂量的双峰驼自然发酵乳一定时期后,较为系统地检测了受试小鼠的抗应激和抗氧化相关指标。在抗应激试验中,将实验动物随机分成3组,Ⅰ组为空白对照组,按0.3 mL /10g剂量灌服生理盐水;Ⅱ组为发酵驼乳低剂量组,Ⅲ组为发酵驼乳高剂量组,分别按0.15 mL /10g剂量和0.3 mL /10g剂量灌服双峰驼自然发酵乳,连续灌服7天,观察双峰驼自然发酵乳对小鼠负重游泳时间﹑耐常压缺氧时间﹑耐高温时间和耐低温时间的影响;定量检测对慢性热应激小鼠的日增重、饲料转化率、胸腺指数和脾脏指数以及血清葡萄糖、白蛋白和总蛋白含量、碱性磷酸酶(AKP)活力、肌酸激酶(CK)活力、乳酸脱氢酶(LDH)活力等的影响。抗氧化试验的分组、灌服发酵驼乳的途径和剂量同上。在末次灌服30min后,采血并处死取样,测定血清、肝、脑和骨骼肌的总超氧化物歧化酶(T-SOD)、过氧化氢酶(CAT)、谷胱甘肽过氧化物酶(GSH-Px)活力,丙二醛(MDA)含量和总抗氧化能力(T-AOC)等。试验结果显示,双峰驼自然发酵乳高剂量组能显著延长小鼠负重游泳时间﹑耐缺氧存活时间﹑耐高温存活时间、耐低温存活时间,极显著提高血清葡萄糖、总蛋白含量和极显著降低AKP、CK活性;白蛋白含量和LDH活性有所上升,但尚未达到显著水平;双峰驼自然发酵乳高剂量组还能显著或极显著(p<0.05或p<0.01)降低小鼠血清、肝、脑组织和骨骼肌MDA含量;能显著或极显著(p<0.05或p<0.01)提高血清、脑组织和骨骼肌T-SOD、CAT、GSH-Px和T-AOC活力,而对肝组织T-SOD、CAT、GSH-PX和T-AOC活力的影响则无明显的规律性。因此,双峰驼自然发酵乳具有一定的抗急慢性应激反应作用和较强的抗氧化作用。

【Abstract】 To investigate the effects of bactrian camel fermented milk on the anti-stress and antioxidant capacity of mice, the some related indicators of anti-stress and antioxidant were systematically evaluated on kunming mice administered with different doses of camel milk in this study. In anti-stress experiment: the experimental animals were randomly allocated into 3 group of groupⅠ,ⅡandⅢ. GroupⅠwas the control, received 0.3 mL / 10g dose of normal saline; GroupⅡa ndⅢwere experimental group administered by per os with 0.15 mL/10g and 0.3 mL/10g of bactrian camel fermented milk respectively for 7 successive days. Then the effects of fermented camel milk on the time for loading swimming, hypoxia survival, heat resisting survival and cold resisting survival of mice were detected. As well as, the effects of fermented camel milk on the daily weight gain, feed conversion rate, thymus and spleen index, the content of serum glucose, albumin and total protein, the activity of alkaline phosphates (AKP), creatine kinase (CK) and lactate dehydrogenises (LDH) were examined in chronic heat stressed mice, respectively. In Antioxidation experiment: the dose and administration route of fermented camel milk were same as anti-stress experiment. After 30min of last administration, blood samples were collected and the activity of total superoxide dismutase(T-SOD), catalase (CAT), glutathione peroxidase (GST-PX), total antioxidation capacity (T-AOC) and the content of malondialdehyde (MDA) in liver, muscle and brain tissue were detected respectively.The result showed that Bactrian camel fermented milk in high-dose group significantly extended the time of loading swimming, hypoxia survival, heat resisting survival of mice, and significantly increased the content of serum total protein and glucose and significantly decreased the activity of AKP and CK. Compared with the control group, the high dose of the Bactrian camel fermented milk significantly decreased the activities of GSH-PX, CAT and T-AOC and the content of MDA in serum, liver, muscle and brain tissue, respectively; significantly increased the activities of T-SOD, GSH-PX, CAT and T-AOC in serum, muscle and brain tissue, respectively. However, there were no significant regularity for the activities of T-SOD, CAT and T-AOC in liver at the high dose of the Bactrian camel fermented milk group. Therefore, Bactrian camel fermented milk has a certain effects on anti-stress and antioxidant capacity of mice.
