补肾活血对大鼠慢性高眼压模型RGCs凋亡基因Bcl-2、Bax表达及多焦视网膜电图的影响

【摘要】 目的:观察补肾活血中药对SD大鼠慢性高眼压(elevated intraocular pressre, EIOP)模型视网膜神经节细胞凋亡基因Bcl-2、Bax表达及多焦视网膜电图的影响,并对其作用机理进行初步探讨。方法:采用烙闭上巩膜静脉法,烙闭大鼠3支上巩膜静脉,制作大鼠慢性EIOP模型,随机分为5组:模型对照组,补肾活血高剂量组、中剂量组、低剂量组,空白对照组。连续灌胃8周,并于8周末处死大鼠,观察其对EIOP大鼠眼压(intraocular pressre, IOP)、多焦视网膜电图(multifocal electroretinogram, mfERG)和视网膜神经节细胞(retinal ganglion cells, RGCs)凋亡基因的影响。结果:本实验采用的烙闭上巩膜静脉的造模方法使大鼠IOP明显升高,在造模后8周(实验结束)IOP仍为造模前的3倍;补肾活血对大鼠慢性EIOP模型视功能损害的干预作用如下:轻度降低IOP,IOP下降约5 - 8mmHg,以高剂量组最明显;有效改善EIOP大鼠mfERG,恢复P1、N1波的反应密度,缩短P1波的峰潜时;调控视网膜神经节细胞凋亡基因的表达,表现为上调抗凋亡基因Bcl-2,抑制凋亡促进基因Bax。结论:采用烙闭上巩膜静脉方法建立的慢性EIOP大鼠模型可获得不少于两个月的中等度EIOP;补肾活血可以有效地保护EIOP模型视功能,其作用主要表现在轻度降低IOP,改善mfERG,上调视网膜神经节细胞抗调亡基因Bcl-2、抑制凋亡促进基因Bax。

【Abstract】 Objective:To observe the effect of traditional Chinese medicine(TCM) of BuShenHuoXue on retinal ganglion cells (RGCs) apoptosis gene Bcl-2, Bax expression and multifocal electroretinogram (mfERG) in rat model of elevated intraocular pressre (EIOP)and to explore the mechanism of it initially.Methods:By unilaterally cauterizing 3 episcleral vessels,the rat model of EIOP was gotten. Rats were divided into 5 groups randomly:model control group, treatment groups with high, middle, low dosage of BuShenHuoXue of TCM, and normal control group.After given drugs or normal saline for 8 weeks, the rats were put to death and eyeballs were taken out. To observe the effect by intraocular pressre(IOP), multifocal electroretinogram(mfERG),retinal ganglion cells(RGCs) apoptosis-related gene.Results:Elevated intraocular pressre was gotten and there was an approximately 3-fold increase in IOP for 8 weeks comparing to premodels building. BuShenHuoXue of TCM can lower IOP slightly, IOP decreased by about 5-8mmHg, with the most obvious high-dose group,and improve mfERG, recover the reaction density of P1, N1 wave, shorten the P1 wave peak latency, regulate the expression of RGCs apoptosis-related gene by increaseing anti-apoptosis Bcl-2 gene expression and lowering the apoptosis inhibitory gene Bax expression.Conclusion:This model moderately elevated IOP can last at least 2 months.BuShenHuoXue of TCM can protect the visual function from the rat model of EIOP, by improving mfERG, increaseing anti-retinal apoptosis Bcl-2 gene expression and lowering the apoptosis inhibitory gene Bax expression.
