藏药昌都锦鸡儿化学成分的研究

【摘要】 昌都锦鸡儿(Caragana changduensis)是豆科锦鸡儿属植物,灌木,主产于我国西藏、青海。昌都锦鸡儿长期以来作为藏药藏锦鸡儿的主要植物来源之一,藏文名为“佐木兴”,其根茎具有活血化瘀、降压、消炎散肿等功效,内用治疗高血压、多血症、月经不调,外用治疗跌打损伤等症。目前,该植物化学成分及药理作用尚无文献报道。本课题开展了昌都锦鸡儿化学成分和药理活性研究,实验采用硅胶、聚酰胺、凝胶等色谱方法对昌都锦鸡儿根茎的木部心材乙醇提取物进行了系统分离,得到15种单体化合物,通过紫外光谱、红外光谱、质谱、核磁共振等手段鉴定了其中12个化合物的结构,分别为10个异黄酮:樱黄素(CaC-2)、阿夫罗摩辛(CaC-3)、3-甲氧基奥洛波尔(CaC-4)、芒柄花素(CaC-5)、odoratin (CaC-7)、木豆素(CaC-8)、毛蕊异黄酮(CaC-11)、染料木素(CaC-13)、5,7,2’,4’-四羟基-5’-甲氧基异黄酮(CaC-14)、兰花赝靛素(CaC-15);1个查尔酮:异甘草素(CaC-10);一个二氢黄酮:甘草素(CaC-12);化合物CaC-1、CaC-6、CaC-9正在结构鉴定之中。其中CaC-2、CaC-4、CaC-13、CaC-14为锦鸡儿属植物中首次分离出的化合物。利用体外测定大鼠巨噬细胞NO生成量的活性筛选方法测定了化合物的活性,其中化合物CaC-11和CaC-13具有很强的抑制大鼠巨噬细胞NO生成的作用。本论文研究成果为阐明藏药藏锦鸡儿的药效物质基础及作用原理提供了科学依据,为该类藏药的进一步开发奠定了基础。
【Abstract】 Caragana changduensis is a shrub of genus Caragana Fabra.,Leguminosae,mainly distributed in Tibet and Qinghai province. C. Changduensis of which Tibetan name is zuomuxing, has a profound medicinal history in Tibetan traditional medicine. Its rhizome has the efficacy of promoting blood flow, reducing blood pressure and eliminating inflammation, which can be treated with plethora, hypertension, catamenia disorder for internal use while bruises and contused wound for external use. Until now, there hasn’t been any reports about its chemical constituents and pharmacological action. Study on chemical constituents will benefit explaining the material basis of the drug. Hence, systematic isolation of ethanol extract of C. Changduensis was carried out. Fifteen compounds were obtained by preparative chromatography methods of silica gel, polyamide, sephadex, toyopearl, et al. Based on UV, IR, MS and NMR, of which twelve were identified as fallows, 10 isoflavones: prunetin(CaC-2), afrormosin (CaC-3), 3’-O-Methylorobol (CaC-4), formononetin (CaC-5), odoratin (CaC-7), cajanin (CaC-8), calycosin(CaC-11), genistein(CaC-13), 5,7,2’,4’-tetrahydroxy-5’-methoxyisoflavone(CaC-14), taxasin (CaC-15), 1 chalcone: isoliquiritigenin (CaC-10); 1 flavanonol: liquiritigenin (CaC-12). CaC-1、CaC-6、CaC-9 are being identified, while CaC-2、CaC-4、CaC-13、CaC-14are isolated from genus Caragana for the first time. The inhibitory effects of these compounds on NO production in lipopolysaccharide (LPS) and interferon-γ(IFN-γ) activated murine macrophage-like cell line (RAW264.7) were tested by microplate ultraviolet-colorimetric method. Compound CaC-11 and CaC-13 showed strong inhibitory effects on NO production.The study results of the paper provide a scientific basic of pharmacodynamic material foundation and action principle of Tibetan Medicine Caragana, and lay a foundation of futher development of such Tibetan Medicine.
