针刺四神聪穴对失眠大鼠下丘脑内5-HT和5-HIAA含量的影响

【摘要】 目的:1.观察针刺四神聪穴对大鼠自主活动、直接入睡率、戊巴比妥钠所致睡眠时间及戊巴比妥钠阈下剂量所致大鼠入睡率变化,明确针刺四神聪穴改善失眠症的疗效。2.观察针刺四神聪穴对失眠大鼠下丘脑内5-HT和5-HIAA含量的变化,揭示5-HT和5-HIAA参与针刺疗效的中枢机制。方法:1.取雄性Wistar大鼠30只,随机分为空白对照组、手针组、电针组,每组10只。使用SD-5G型药理生理实验多用仪记录针刺后各组大鼠的自主活动变化。2.分别取雄性Wistar大鼠30只,随机分为空白对照组、手针组、电针组,每组10只,分别观察针刺后大鼠的直接入睡率、戊巴比妥钠所致大鼠的睡眠时间变化、戊巴比妥钠阈下剂量所致大鼠的入睡率变化。3.选取雄性Wistar大鼠50只,随机分成5组:空白对照组(简称空白组)、模型对照组(简称模型组)、安定组、手针组、电针组、采用腹腔注射对氯苯丙胺酸(PCPA)制备失眠动物模型,通过7日针刺及药物组安定灌胃后,取大鼠下丘脑,通过荧光分光度法测定下丘脑内5-HT和5-HIAA含量。结果:1.手针组及电针组均可减少大鼠的自主活动,与对照组比较有显著性差异(P<0.01)。电针组与手针组相比,减少大鼠的自主活动更为明显,两组比较有显著性差异(P<0.01)。2.手针组及电针组大鼠的直接入睡率为零。手针组及电针组可以延长戊巴比妥钠所致大鼠睡眠时间,与对照组比较,有显著性差异(P<0.01);电针组与手针组比较,有显著性差异(P<0.01),即电针可以明显延长戊巴比妥钠所致的大鼠睡眠时间。手针及电针四神聪穴可以增加戊巴比妥钠阈下剂量所致大鼠入睡率,与对照组比较,有显著性差异(P<0.01);电针组与手针组比较,有显著性差异(P<0.01)。3.PCPA腹腔注射后大鼠正常的睡眠-觉醒周期消失,且大鼠下丘脑内5-HT、5-HIAA的含量下降,与空白组比较有统计学意义(P<0.05)。模型组与空白组比较,下丘脑中5-HT和5-HIAA含量明显低于空白组,二者有显著性差异(P<0.01)。安定组与模型组相比,下丘脑中5-HT和5-HIAA含量无明显变化,二者无显著性差异(P>0.05)。手针组及电针组下丘脑中5-HT和5-HIAA含量均增加,与模型组相比,有显著性差异(P<0.01),且与安定组相比,有显著性差异(P<0.01)。电针组同手针组相比,下丘脑中5-HT和5-HIAA含量增加明显,二者有统计学意义(P<0.05)。结论:1.针刺四神聪穴可减少大鼠的自主活动、延长戊巴比妥钠所致大鼠睡眠时间及增加戊巴比妥钠阈下剂量所致大鼠入睡率,提示针刺四神聪穴对大鼠有镇静催眠作用,且以电针作用为明显。2.针刺四神聪穴对大鼠没有明显的直接睡眠作用。3.针刺四神聪穴可使PCPA化失眠大鼠下丘脑内降低的5-HT、5-HIAA含量升高,以电针作用为明显,这可能是实现其增加睡眠,提高睡眠质量的作用机制之一。

【Abstract】 Objective: 1. To observe the influence of spontaneous activity direct sleep rate the sleeping time of rats eoursed by sodium pentobarbltal and sleep rate eoursed by sodium pentobarbltal at the critical dose , explicit the effection of insomnia by Sishencong acupuncture. 2. To observe the content of 5-HT and 5-HIAA in the insomnia rats’ Hypothalamus caused by Sishencong acupuncture, thus to reveal the central mechanism of the treatment for insomnia by acupuncture.Methods: 1. Thirty male Wistar rats are randomly divided into three groups: blank control group (n=10); hand acupuncture group (n=10); electro-acupuncture group (n=10 ) .By using SD-5G physiological and pharmacolo-gical experimental equipment record the spontaneous activity of each group after acupuncture . 2. Thirty male Wistar rats are respectively and randomly divided into three groups : blank control group (n=10); hand acupuncture group (n=10); electro-acupuncture group (n=10) .Observe the influence of spontaneous activity direct sleep rate the sleeping time of rats eoursed by sodium pentobarbltal and sleep rate eoursed by sodium pentobarbltal at the critical dose respectively.3.Fifty male Wistar rats are divided into five groups at random: Blank control group (n=10); Model control group (n=10); Diazepam group (n=10) .Hand acupuncture group(n=10); Electro-acupun- cture group(n=10); Duplicate insomnia model of rats by means of PCPA abdominal injection. The animals in the two acupuncture groups will be treated with 7 days’ acupuncture, the animals in Diazepam group will be treated with seven days’ diazepam intragastric administration, then study the results of the treatments by observing the content of 5-HT, 5-HIAA in the insomnia rats’ Hypothalamus.Results: 1. Both hand acupuncture group and electro-acupunc- ture group can ruduce the spontaneous activity with comparison to control group(P<0.01), electro-acupuncture group reduce the spontaneous activity more obviously compared with hand acupuncture group (P<0.01) .2.There is no direct sleep rate both in hand acupunc- ture group and electro-acupuncture group. Hand acupuncture group and electro-acupuncture group can prolong the sleeping time of rats eoursed by sodium pentobarbltal and increase sleep rate eoursed by sodium pentobarbltal at the critical dose, with significance difference comparison to control group ( P<0.01 ) .Compared with hand acupuncture group, electro-acupuncture group has significance difference (P<0.01) . 3.After PCPA abdominal injection the normal sleep of rats disappeared, at the same time, the content of 5-HT and 5-HIAA in the Hypothalamus of rats decreased with comparison to blank control group (P< 0.05) . Model group compared with the blank, the content of 5-HT and 5-HIAA in the Hypothalamus of rats decreased obviously, has significance difference ( P<0.01). There is no significant change between Diazepam group and Model control group ( P>0.05) . Both hand acupuncture group and electro-acupuncture group can increase the content of 5-HT, 5-HIAA in the insomnia rats’ Hypothalamus, with significance difference comparison to model control group (P<0.05), and have significance difference comparison to Diazepam group (P<0.01) . Compared with hand acupuncture group, electro-acupuncture group has signifi-cance difference (P<0.05) .Conclusions : 1. Sishencong acupuncture can ruduce the spontaneous activity, prolong the sleeping time of rats eoursed by sodium pentobarbltal and increase sleep rate eoursed by sodium pentobarbltal at the critical dose. Sishencong acupuncture has sedative and hypnotic effects, espencialy electro-acupuncture.2.Sishencong acupuncture has no obviously direct sleep effects on rats.3.Acupuncture can increase the content of 5-HT, 5-HIAA in the insomnia rats’ Hypothalamus. This may be one of the mechanisms that acupuncture can promote sleep and enhance the quality of it.
