复元胶囊对气虚血瘀证大鼠血液流变学和NO、TXB2、ET、6-keto-PGF1a的影响

【摘要】 目的:(1)研究气虚血瘀证大鼠血液流变学及血清一氧化氮(NO)、血浆内皮素(ET)、血栓素B2(TXB2)、6-酮-前列腺素F1a(6-keto-PGF1a)的变化。(2)研究中药复元胶囊对气虚血瘀证大鼠血液流变学及血清一氧化氮(NO)、血浆内皮素(ET)、血栓素B2(TXB2)、6-酮-前列腺素F1a(6-keto-PGF1a)的影响。方法:(1)选用健康18月龄雌雄各半SD大鼠72只,随机分为造模组和空白对照组,以饥饿、寒冷、疲劳及高脂饮食诱导大鼠气虚血瘀证模型。(2)造模成功后随机将造模组分为模型组、阳性对照组、复元胶囊大剂量组、复元胶囊中剂量组、复元胶囊小剂量组,每组12只。(3)各复元胶囊组分别给予复元胶囊灌胃;阳性对照组给予芪参胶囊灌胃;模型组和空白对照组给予等量生理盐水灌胃。(4)4周后测定大鼠的耐疲劳时间,双球直管式检测血液流变学,硝酸还原酶法检测NO,放射免疫法(RAI)检测TXB2、ET及6-keto-PGF1a。结果:(1)耐疲劳时间模型组与空白组比较明显缩短(P<0.01)。(2)血液流变学模型组高于空白对照组(P<0.05)。(3)血清NO、血浆6-keto-PGF1a模型组低于空白对照组(P<0.05);而ET、TXB2高于空白对照组(P<0.05)。(4) TXB2分别与NO、6-keto-PGF1a呈显著负相关(P<0.001);而与ET呈显著正相关(P<0.001)。(5)耐疲劳时间复元胶囊大、中剂量组比复元胶囊小剂量组及芪参胶囊组显著延长(P<0.01)。(6)血清NO、血浆6-keto-PGF1a复元胶囊中剂量组比复元胶囊大剂量组、复元胶囊小剂量组及芪参组增高(P<0.05);而ET、TXB2降低(P<0.05)。(7)血液流变学复元胶囊中剂量组比复元胶囊大剂量组、复元胶囊小剂量组及芪参组低(P<0.05)。结论:(1)气虚血瘀证大鼠血液流变学存在浓、粘、凝、聚的状态。(2)气虚血瘀证大鼠NO/ET、6-keto-PGF1a/TXB2比值严重失调。NO、6-keto-PGF1a显著降低,ET、TXB2显著升高。反映了气虚血瘀证大鼠存在着前列腺素代谢失调,NO、ET合成或分泌失衡。(3)中药复元胶囊可改善气虚血瘀证大鼠血液流变学、NO/ET、6-keto-PGF1a/TXB2比值,升高NO、6-keto-PGF1a水平;降低ET、TXB2水平,且疗效优于芪参胶囊,这可能与复元胶囊有效改善VEC的结构和功能有关。

【Abstract】 Objective(1)To investigate the changes of blood viscosities、serum NO、plasma ET、TXB2、6-keto-PGF1a of rats with qi-deficiency and blood stasis syndrome.(2)To investigate the mechanisms of the fuyuan capsule effect on the changes.Methods(1)18-month-old healthy male and female Sprague-Dawley(SD) 72 rats were divided into 2 groups randomly: model group and blank control group,rats were induced by exertion, starvation, cold and high fat food , forming models with Qi Deficiency and Blood Stasis syndrome.(2)rats with model group appear the syndrome of qi deficiency、blood stasis and rise of blood viscosities. rats were divided into 6 groups randomly and equally: model group, positive control group (Qi-shen capsule), Fu-yuan capsule large dose group, Fu-yuan capsule middle dose group , Fu-yuan capsule little dose group , blank control group,every group has 12 rats.(3)Fu-yuan capsule group and positive control group (Qi-shen capsule) were respectively fed with Fuyuan capsule and Qishen capsule, and model contrast group and blank control group were fed with 0.9 N.S at the same time.(4)The time about fatigue-resistant were measured ,blood viscosities were measured by revolve viscosities apparatus, ET、6-keto-PGF1αand NO level are measured by Radioimmunoassay after 4 weeks.Results:(1)the time fatigue-resistant in model group were significantly shorter than those in blank control group(P<0.01).(2)the level of blood viscosities in model group were higher than those in blank control group(P<0.05).(3)the level of NO、6-keto-PGF1αin model group were significantly lower than those in blank control group(P<0.05),but the levels of ET and TXB2 in model group were significantly higher than those in blank group(P<0.05).(4)there were significantly negative correlation between the levels of TXB2 and 6-keto-PGF1αin blood serum or TXB2 and NO in blood serum(P<0.001);however, there were significantly positive correlation between the level of TXB2 and ET(P<0.001). (5) compared with Fuyuan capsule little dose group、Qishen capsule group, the time fatigue-resistant of rats were lengthened in Fuyuan capsule large dose group and Fuyuan capsule middle dose group(P<0.05).(6) compared with Fuyuan capsule large dose group、Fuyuan capsule little dose group、Qishen capsule group ,the levels of NO and 6-keto-PGF1αwere significantly raised in Fuyuan capsule middle dose group(P<0.05),but the levels of ET and TXB2 were descended(P<0.05).(7)compared with other treatment groups, the levels of blood viscosities were comprehensively reduced in Fuyuan middle dose group (P<0.05).Conclusions(1)The blood viscosities about rats who were suffering from Qi-deficiencyand blood stasis syndrome was having keeped concentrated、sticky、congelative、assemblily state.(2)The ratio of NO/ET and TXB2/ 6-keto-PGF1αtook place seriously maladjustment,NO and 6-keto-PGF1αlevels markedly decresed,but TXB2 and ET levels remarkablely increased;these facts can reflect the complex situation of maladjustment from prostaglandins metabolism and the unbalance synthesis and secretion of NO and ET.The above factors could relate to VEC damnification.( 3 ) Fuyuan capsule can effectively ameliorate the serious maladjustment of ratio of NO/ET and TXB2/6-keto-PGF1α,raise the levels of NO and 6-keto-PGF1a in blood,meanwhile reduce the levels of ET and TXB2,the above effect of Fuyuan capsule was better than Qishen capsule.the phenomena of Fuyuan capsule could relate to its abilities to improvement of the function and renew of configuration for VEC.
