南蛇藤生药学及指纹图谱研究

【摘要】 南蛇藤为卫矛科南蛇藤属植物南蛇藤(Celastrus orbiculatus Thunb.),为民间习用药材。具有驱风除湿,活血解毒消肿的功效,民间常用该药煎汤或外敷治疗风湿痹痛。现代药理研究表明其具有抗炎、抗肿瘤、免疫抑制等多种生物活性,临床上可用于治疗类风湿性关节炎和多种自身免疫性疾病。该植物分布广泛,资源丰富。与它同科的雷公藤和昆明山海棠是有毒植物,三者毒性不一,并在市面上有混淆的现象,若不加以区分,将影响临床用药安全。本文对南蛇藤有效成分、现代药理研究、临床作用以及应用中呈现的毒副作用做了综述和总结,对其生药学和主要有效成分进行研究,初步建立了其质量控制方法。本文应用显微、薄层色谱、高效液相色谱、红外光谱等技术对该南蛇藤进行性状特征、显微鉴定、理化鉴别等方面的生药学研究。其中显微鉴定包括横切面和粉末的鉴定;理化鉴别包括薄层色谱、液相指纹图谱和红外指纹图谱。药材鉴别可大致反映药材的真伪优劣,是中药鉴定的可靠依据,本文研究了南蛇藤性状和显微特征,有助于区分南蛇藤正品和混淆品。本文对南蛇藤中的主要有效成分—卫矛醇和扁蒴藤素进行了研究,经过实验分别建立了卫矛醇和扁蒴藤素的薄层鉴别法。此法简便稳定,为南蛇藤质量标准的制定提供参考。本文采用高效液相色谱和蒸发光散射检测器联用技术,建立了卫矛醇的含量测定法。本方法操作简单,结果稳定,可作为南蛇藤类药材及其制剂中卫矛醇的定量分析方法。我们测定了不同产地的南蛇藤中卫矛醇的含量,发现各地的差异很大,说明当前南蛇藤药材的质量控制需要加强。在含量测定研究的基础上,我们还对南蛇藤进行了指纹图谱方面的研究。以扁蒴藤素为指标成分,建立了南蛇藤的高效液相指纹图谱。经方法学考察,该法稳定可靠、专属性强。在研究整体图谱的基础上,通过参照峰与其余十四个共有色谱峰的相对面积实现南蛇藤的质量控制。本次研究也为南蛇藤有效组分与药效之间的关系-即组效关系的研究奠定了基础。我们测定了不同产地的南蛇藤指纹图谱,发现药材的地区差异很大,进一步说明当前南蛇藤药材的质量控制需要加强。本文用红外光谱法比较卫矛科的南蛇藤和昆明山海棠的粉末及醇提物的特征。两种药材的粉末图谱中都看到有草酸钙和淀粉的特征峰,醇提物图谱中都能看到卫矛醇的特征峰,但南蛇藤较为明显。从两种药材醇提物的二阶导数图谱中可看到南蛇藤与昆明山海棠有明显差异,说明它们某些成分含量不同。南蛇藤根图谱的草酸钙和卫矛醇的峰形都比茎的强和尖,说明根中草酸钙和卫矛醇的含量比茎高。由于红外法既能快速地找出药材差异,又能准确指认出卫矛醇,所以此法制定的宏观指纹图谱可作为鉴定南蛇藤的简易方法。

【Abstract】 Celastrus orbiculatus Thunb. is one of folk medicine and distributes widely in China. It was used for activating blood, detoxification and detumescence.The extracts of Celastrus orbiculatus Thunb. have various activities, inculding anti-inflammatory, anti-tumorand, immunosuppressive, etc. Its preparation was used for the treatment of rheumatoid arthritis, and other autoimmune diseases in clinical.Celastrus orbiculatus Thunb. was reviewed in its distribution, application, chemical components, pharmacological function, clinical applications, etc. And also, its pharmacognostic properties, main contents and quality control were studied.In this thesis, both dulcitol and pristimerin, the primary contents in Celastrus orbiculatus Thunb., have been studied by Thin Layer Chromatography (TLC). The method may help to establish the quality standards for Celastrus orbiculatus Thunb. and its preparation.The method of content determination was established by chromatographic experiments. The results showed the method was easy to operate and accurate. Content of dulcitol from Celastrus orbiculatus Thunb. was determined by HPLC method. The results showed that the contents of dulcitol from Celastrus orbiculatus Thunb. in different producing area revealed a sharp difference, and this result indicated the quality control of Celastrus orbiculatus Thunb. should be improved.The Chromatographic Fingerprint of its bioactive component was established by HPLC method. The methods were stable, reliable and specific for analysis bioactive component from Celastrus orbiculatus Thunb. The method can be applied to the quality control of Celastrus orbiculatus Thunb. In addition, as a further research, the relationship between the bioactive components group and their effects, was necessary and important.Moreover,The FTIR spectra of Tripterygium hypoglaucum (level) Hutch (THH) and Celastrus orbiculatus Thunb are studied. The result show that the two herbs have different ratio of main substance calcium oxalate and dulcitol, so their absorption frequency and form are different as well. There is an obvious different between Celastrus orbiculatus stem and root,both the peak form of calcium oxalate and dulcitol in the FTIR spectra of roots are sharper and stronger than stems. Therefore FTIR can be a fast and simple method to indentify Celastrus orbiculatus.
