五种药用植物生长过程中化学成分的变化及天麻素苷元衍生物的合成研究

【摘要】 本论文分两个部分,第一部分五种药用植物在不同生长时期中主要化学成分的变化研究;第二部分是天麻素苷元及其衍生物的合成研究。植物次生代谢产物会随着其生长过程而呈动态变化,分析研究不同时间段的次生代谢产物有可能揭示其动态变化过程,并为揭示次生代谢产物的生物合成提供依据。本部分实验以女贞子、辛夷、连翘、山茱萸、忍冬等为研究对象,应用高效液相色谱法,分析其不同生长时期的花蕾、花、叶、果实等的化学成分,揭示其化学成分的变化过程。结果显示这五种药用植物的花蕾、花、叶、果实整个生长过程中主要化学成分存在此消彼长的动态变化,有些成分在其生长过程中存在,在干品已经消失。天麻素是名贵药材天麻的有效单体,据现代药理研究,它具有镇静、抗惊厥、抗癫痫等多种功能。天麻素主要来源于天然产物中,并且含量较低,一般情况下低于百分之一。直接从天然产物中提取成本很高,人们期望依靠化学合成的方法获取价廉的化合物为人类所用。本文共合成出十六个天麻素苷元衍生物,意图通过活性检测,筛选出具有良好生物活性的化合物。

【Abstract】 This dissertation is divided into two parts. The first part is the study of chemical profile of five medicinal plants during the growth period. The second part is the synthetic study of aglycone of gastrodin and its derivatives.To study the distribution and dynamic change of chemical components in the fresh and dry buds, flowers, leaves and fruits of medicinal plants, the fresh and dry samples were extracted with methanol and analyzed by HPLC respectively. The chromatograms of fresh and dry samples were obtained and compared. There is a dynamic change of major components in medicinal plants during the growth process. There is also a postharvest process in some plants after fresh sample cut off.Gastrodin is an active constituent in traditional Chinese medicine Gastrodia elata Bl. According to modern pharmacological research, it has many biological activities, such as sedative, anticonvulsant, epilepsy, and so on. So far, gastrodin comes mainly from natural plant, and content is too low, generally lower than the 1.0%. It has a strong demand of synthetic aglycone of gastrodin and its derivatives. 16 gastrodin aglycone derivatives were prepared with simple methods in this paper.
