西黄丸含药血清抗人乳腺癌和人卵巢癌作用的研究

【摘要】 西黄丸,又名犀黄丸,方出清—王洪绪所著《外科证治全生集》,为治疗“乳岩”、“瘰疬”、“痰核”、“肺痈”之名方。现代药理学研究表明,西黄丸具有抗肿瘤、增强免疫功能、抗菌及抗炎作用等活性,临床上主要用于多种恶性肿瘤(如肝癌、白血病、肺癌、乳腺癌、卵巢癌、结肠癌等),尤其适用于晚期恶性肿瘤、和乳腺增生等良性疾病的治疗。在国内中成药抗癌药物中是一个较理想的中药制剂。尽管该方剂临床治疗效果尤其抗肿瘤作用已得到了肯定,但是其抗肿瘤的靶点和其作用的分子机制尚未阐明。本研究选择两种严重威胁妇女健康的恶性肿瘤——乳腺癌和卵巢癌瘤株,采用血清药理学方法通过体外肿瘤细胞培养,以肿瘤细胞抑制率为指标,观察西黄丸含药血清对人乳腺癌和人卵巢癌两种肿瘤细胞生长抑制作用以及细胞周期的影响。力求从细胞水平探讨其抗乳腺癌和卵巢癌细胞的作用机制。我们将西黄丸制成水溶液,按一定的剂量给CV级SD大鼠灌胃。按照血清药理学实验方法,获取西黄丸含药血清,将含药血清稀释成几种浓度。取人乳腺癌和卵巢癌细胞传代培养,待细胞培养成功后,将几种浓度的血清分别作用于人乳腺癌和卵巢癌细胞细胞。培养24小时和48小时后,用MTT比色法,测各组细胞的光密度值(OD值)计算癌细胞生长抑制率。流式细胞术,测定各组细胞的细胞凋亡率并进行细胞周期分析。实验结果显示西黄丸含药血清对人乳腺癌细胞系(MCF-7)有较强抑制作用,其抑制率与含药血清浓度线性相关。经西黄丸含药血清处理24小时后,乳腺癌细胞系(MCF-7)S期细胞比例增加,G2/M期细胞减少;处理48h后,G0/1期细胞增多,S期细胞减少。西黄丸含药血清对人卵巢癌细胞(SK-OV-3)也具有较强的抑制作用,其抑制率也与含药血清浓度呈正相关。经西黄丸含药血清处理72h,S期细胞比例增加,G2/M期细胞减少,G0/1期细胞减少。由此得出如此下结论:1.西黄丸能够明显抑制乳腺癌(MCF-7)细胞的增殖,其血清浓度与抑制率呈现正相关。2.西黄丸含药血清可抑制人乳腺癌细胞系(MCF-7)细胞生长,并可干扰其细胞周期。因西黄丸含药血清早期主要影响细胞进入G2/M期,后期主要使细胞停滞于G0/1期。由于大部分细胞停滞在DNA合成前期,抑制了癌细胞的增殖而导致其发生凋亡,因此,推断干扰细胞周期可能是其抗癌作用的重要机制。3.西黄丸能够明显抑制人卵巢癌(SK-OV-3)细胞的增殖,其血清浓度与抑制率呈现正相关。4.西黄丸通过对卵巢癌SK-OV-3细胞周期的显著影响,将其阻滞于S期并抑制其增殖最终导致卵巢癌SK-OV-3细胞的凋亡是其治疗人类卵巢癌的重要机制。

【Abstract】 Objective: To investigate the effects of Xihuang pill drug serum on the growthand the cell cycle of breast carcinoma cell (MCF-7) and ovary carcinoma cell(SK-OV-3). Method: activity of tumor cell was detected by MIT and cell cycle wasanalyzed by flow cytometry. Result: Xihuang pill drug serum had an inhibitory effecton breast carcinoma cell (MCF-7) and, ovary carcinoma cell (SK-OV-3) which waspositively related with the drug content. Typital morphological changes of apoptosiswere observed under the transmission electron microscope. FCM showed that the cellcycle treated with Xihuang pill drug serum for 24h, the ration of cells in S phaseincreased, while that in G2/M phase decreased; after 48h treatment, the ratio of cells inG0/1 phase increased, and S phase decreased. Based on these data, we can concludethat Xihuang pill drug serum can inhibit the growth of breast carcinoma cell(MCF-7)and its cell cycle in vitro.And the typital morphological changes of apoptosis were also observed under thetransmission electron microscope. Cell cycle analyzed by FCM indicated that 72hafter Xihuang pill serum treated, the ration of cells in S phase increased and that inG2/M phase decreased. The conclusion resulted from the data related above is thatXihuang pill serum inhibit the growth and cell cycle of ovary carcinomacell(SK-OV-3) in vitro.Cell cycle analyzed by FCM indicated that 72h after Xihuangpill serum treated, the ration of cells in S phase increased and that in G2/M phasedecreased.
