步长脑心通胶囊对脑血栓栓塞大鼠血浆磷脂水平的影响

【摘要】 目的:观察大鼠脑血栓形成后血浆溶血磷脂酸(LPA)和磷脂酸(PA)浓度的变化,以及步长脑心通胶囊对其影响。方法:采用颈内动脉直接插管注入凝血酶建立大鼠脑血栓栓塞模型,并预防性给予步长脑心通胶囊,观察大鼠脑血栓栓塞后溶血磷脂酸(LPA)和磷脂酸(PA)的水平。具体方法如下:选取健康Wistar大鼠24只,随机分为4组,即正常对照组;实验对照组:药物试验组;假手术组,每组6只。24只大鼠均经腹主动脉取全血约4ml,检测LPA和PA:①实验对照组建立脑血栓栓塞模型(参照张兆岩等的方法并加以简化改进建立大鼠脑血栓栓塞模型),术后3小时采血检测;②药物试验组,术前每日0.48g/kg步长脑心通灌胃,共7天,其余过程同实验对照组;③正常对照组和假手术组也经腹主动脉取全血约4ml,检测LPA和PA;结果:实验对照组血浆LPA水平(12.47±1.65)umol/L与正常对照组(2.41±0.17)umol/L相比有很大程度的增高,药物试验组溶血磷脂酸(LPA)值(4.79±0.62)umol/L显著低于实验对照组(12.47±1.65)umol/L[P<0.05]。实验对照组磷脂酸(PA)水平(10.51±0.98)umol/L较正常对照组(3.11±0.60)umol/L明显增高,即脑血栓大鼠血浆磷脂酸(PA)水平与正常对照组相比有很大程度的增高,而步长脑心通使脑血栓大鼠血浆磷脂酸(PA)水平降低(5.71±0.63)umol/L[P<0.05];结论:大鼠脑血栓栓塞后血浆LPA和PA水平有不同程度增高,预防性给予脑心通胶囊可降低LPA水平,同时还可抑制血浆PA水平的增高。步长脑心通胶囊能够从多途径、多环节、多靶点阻断血栓形成,组方中的水蛭素是直接凝血酶抑制剂,丹参有ADP受体拮抗剂类似的生物效应,乳香、红花具有抑制血小板聚集,降低血液粘滞度的作用,全蝎、地龙有抗凝促纤溶作用,当归的水溶性成分阿魏酸能稳定血小板膜,抑制血小板环氧化酶活性,使血栓烷A2(TXA2)降低。该药对血液和血管双重奏效,在抗凝、降纤、溶栓方面发挥了重要作用,从而对血浆LPA和PA值起到修正作用。

【Abstract】 Aim;To investigate the changes of the plasma Phospholipids concentration and the effect of Buchang Naoxintong on it after Acute Cerebral Thromboembolism in rats.Methods;twenty-four wistar rats were randomly divided into five groups(n=6 each):normal control,sham-operation,ischema,and Buchang naoxintong pretreatment.An embolus was placed at the origin of the middle cerebral artery(MCA)by injection of autologous single blood clot via an intraluminal catheter placed in the intracranial segment of internal carotid artery.The concentrations of LPA and PA were quantified by ultraviolet spectrophotometry.Results A total of 24 rats were involved in the result analysis:①The concentrations of plasma LPA(12.47±1.65)umol/L and PA(10.51±0.98)umol/L in the ischemia group was higher compared with the normal control group(2.41±0.17)umol/L(p<0.05 )and(3.11±0.60)umol/L respectively.②The concentrations of plasma LPA and PA decreased significantly in Buchang naoxintong pretreatment group.Conclusions:The concentrations of plasma LPA and PA increases differently after cerebral thromboembolism.Pretreatment with naoxintong can decrease the concentration of plasma LPA in rats,and inhibit the increasing of concentration of plasma PA.Buchuang Naoxintong capsule can inhibit the formation of the cerebral infarction from different ways,different links and different targets,the leech component is the direct Thrombase inhibitor,danshen root component had the analogical biological effect of ADP-receptor antagonist,libanotus and Carthamus tinctorius have the effect of platelet aggregation inhibition and blood Viscosity depression.Scorpion and Ailolobophora caliginosa trapezoides had the effect of anticoagulation.etc So,Buchuang Naoxintong capsule can improve the neural function.Reduce brain edema,and protect the neurons in rats with cerebral infarction.The improvement of cerebral infarction has a positive correlation with the use of naoxintong.It can affect the blood and the blood vessel itself and adjust the the concentrations of plasma LPA and PA.
