利用RAPD标记研究百合的遗传多样性

【摘要】 百合(Lilium)是我国的传统花卉,适于做切花、盆花和园林布景。我国是世界百合的分布中心,拥有约全世界一半以上的品种,但是由于各种人为和自然原因使得我国百合资源遭到严重破坏和流失,导致多样性逐年降低。此外,我国在百合的育种方面非常薄弱,目前还没有培育出较知名的百合杂交品种或品系,当前国内商业栽培种球主要依赖于国外进口。本研究利用RAPD技术对采集到的53个百合品种进行遗传多样性的分析及亲缘关系的鉴定,以期为进一步建立百合核心种质以及更好地利用资源打下基础,为我国百合育种工作提供参考。通过研究,取得了以下进展:1.从全国各地收集到了53份百合种质,其中野生种21份,栽培种32份,为百合种质资源研究和保存打下了基础。2.优化了百合RAPD-PCR的循环条件,得出了适合百合RAPD分析的“双退火温度”循环程序:即37℃30s 5个循环和48℃38个循环的PCR扩增。3.用125个随机引物对10份东方百合杂交系栽培种进行了RAPD分析,成功地筛选出了30个适合于百合RAPD分析的随机引物,这些引物可以扩增出丰富清晰的条带。

【Abstract】 Lily is a famous traditional flower in China. It can be used as food, medicine and ornamental plants. China, the center of lily origin, has over half of all lily species all over the world and has a large number of wild species with high genetic diversity. However, a lot of them are in danger of extinction because of natural disasters, pest and so on. While for some reasons, the breeding of lily is undeveloped in China. At present, almost no famous varieties and cultivars were developed in china, and most of the lily bulbs in commercial use are imported.The genetic diversity of 53 wild species and cultivars of lilies were analyzed by RAPD for the purpose of providing bases of establishing lily core germplasm and better utilization of lily resources. Through study, the following progresses have been made:1. 53 lily gerplasm including 21 wild species and 32 cultivars from all over the China was collected and preserved.2. The RAPD-PCR condition for lily has been optimized, and the suitable RAPD condition was the amplification for 5 cycles at 37℃for 30s followed by 38 cycles at 48℃.3. 30 random primers which could amplify polymorphic and clear bands were selected out for RAPD analysis of lily among 125 primers through
