新疆天然食用植物色素资源开发与利用

【摘要】 本文以新疆地产黑果小檗(Berberis heteropoda Schrenk)果实为试验材料,采用植物学、植物化学、食品卫生学、卫生毒理学的研究方法。采用NKA-9大孔树脂吸附-解吸附方法纯化黑果小檗色素:对该色素进行了定性识别;对色素纯化前后对光、热、还原介质、氧化介质、防腐剂的稳定性进行了比较试验;筛选出了一种有效的稳定剂;并从光、热、PH值、还原介质、氧化介质、蔗糖、防腐剂、金属离子等影响因子方面对色素的稳定性及添加稳定剂后的色素的稳定性进行了比较试验;采用原子吸收分光光度法对色素中有害微量原素砷、铅、汞、铜的含量进行检测;对色素的安全性进行了经口急性毒性试验,小鼠骨髓微核试验、小鼠精子畸形试验等毒理学方面的研究。结果表明:大孔树脂吸附-解吸附法纯化黑果小檗色素实验室工艺简单易行,色素回收率高;黑果小檗色素属于花色苷类色素;耐光性好;对热有一定耐受性;对低浓度氧化剂有一定耐受性;耐还原性差;对防腐剂耐性差;适用PH<6条件下使用;蔗糖和大多数金属离子对色素无不良影响,但Fe3+、Fe2+、Sn2+和低浓度pb2+对色素有影响,加入稳定剂后,可提高色素的耐热性、耐氧化性、耐防腐剂能力;经对有害微量元素进行检测和毒理试验表明,该色素安全无毒,可以作为天然食用色素使用。

【Abstract】 This paper, based on a research of the fruit of Berberis heteropoda Schrenk of Xingjiang, attempts to exploit the utility of the pigment of this plant as possible extracted material for pigment products. The research is finished by use of the theories and methods of many subjects, such as botany, plant chemistry, food hygiene and toxicology. The pigment is purified by the method of adsorption-disadsorption by macroporous adsorption resin NKA-9 and is discerned roughly. A kind of stability matter has been selected. And The experiment studies also include the preliminary study of the effects of light, heat, PH value, reductant, oxide, sucrose, preservative and metal ions on the stability of the pigment. The amount of the contents of harm trace elements contained in the pigment, such as Hg, Pb, As, Cu, are determined by the method of atomic absorption spectrum. The edible safety properties are examined by experiments of acute toxicity, bone marrow cell micronucleus, and sperm deformity. The results show that the method of adsorption-disadsorption by macroporous adsorption resin is simple and easy method. The pigment of Berberis heteropoda Schrenk is a type of anchocyanin and it is strong in resistance against light and to some extent, good in enduring heat and low consistence oxide. But it is poor in resistance against reductant and preservative. It can be applied to the condition of PH<6. It is also found that sucrose and most of metal ions do not exert negative effects on the stability of the pigment, except Fe3+, Fe2+and low consistence Pb2+, which have some extent effects. Adding stability matter can improve its stability. The harm trace elements contains are within requirement of the National Standard on food safety, As the lexicological study indicates that the pigment does not have any toxicity and mutagenesis, it can be used as edible pigment.
