环境内分泌干扰物2,4-二氯苯氧乙酸、阿特拉津对血液中正常性激素干扰的研究

【摘要】 环境内分泌干扰物(Environmental Endocrine Disrupting Chemicals,简称EDCs)是指环境中存在的能干扰人类或动物内分泌系统诸环节并导致异常效应的物质。 随着人们对环境内分泌干扰物研究的深入,越来越多的化学物质被发现具有环境内分泌干扰物的作用。为了深入了解环境内分泌干扰物的危害,并能有效的加以控制,环境内分泌干扰物与生物体的相互作用机理研究已经成为目前的研究热点。针对目前国内在该领域的研究空白,实验选择了两种除草剂对环境内分泌干扰物与动物体血液组分之间的相互作用机理进行了系统研究。 雌二醇、睾酮是两种天然激素,而2,4-二氯苯氧乙酸(2,4-D)和阿特拉津(Atrazine)是已被确认为环境内分泌干扰物的两种农药。实验采用荧光猝灭法分别系统研究了雌二醇、睪酮、2,4-D、Atrazine与牛血清白蛋白、人血清之间的相互作用,发现雌二醇、睾酮、2,4-D、Atrazine能导致牛血清白蛋白和人血清发生结合并导致静态荧光猝灭,在此基础上利用荧光猝灭公式计算了各体系的结合常数以及结合位点,并对比了二元体系中环境内分泌干扰物与天然激素作用的不同。 为了进一步揭示环境内分泌干扰物在血液系统中对生物体产生影响的作用机理,实验系统研究了天然激素/环境内分泌干扰物/牛血清白蛋白(人血清)三元体系中天然激素与环境内分泌干扰物之间的竞争结合作用,利用双倒数公式计算了不同浓度的环境内分泌干扰物或天然激素与牛血清白蛋白、人血清的结合常数,得出了三元体系中环境内分泌干扰物或天然激素浓度对结合常数的影响,并由此推测了血液系统中环境内分泌干扰物对血液中激素平衡可能具有的干扰机理。

【Abstract】 Environmental Endocrine Disrupting Chemicals (EDCs) is defined as an exogenous substance that presents in the environment and causes adverse health effects in an intact organism, or its progeny, consequent to changes in endocrine function.With the development of EDCs research, more and more chemical materials are considered as "potential endocrine disrupter" which might be expected to lead to endocrine disruption in an intact organism. In order to develop the knowledge of EDCs’ harm and control it more efficiently, research of the interaction mechanism between EDCs and organism has been valued by more and more researchers.Estradiol, testosterone are two hormones in human body, and 2,4-D , Atrazine are considered as two kind of EDCs. The interaction between estradiol, testosterone, 2,4-D, Atrazine in BSA or human serum were studied by fluorescence analysis. The results show that both hormones and EDCs have a strong ability to quench BSA or human serum fluorescence, which is mainly through a static quench procedure. The binding constant K and the number of binding site n were calculated according to the fluorescence quenching expressions, and the difference between hormones and EDCs in organism was compared.In order to develop the interaction mechanism between EDCs and organism, the interaction between hormone and EDCs in hormone/EDCs/BSA (human serum) ternary system was investigated by fluorescence measurement. The binding constant K was calculated with Lineweaver-Burk method, and the influence between hormone and EDCs on binding constant K was obtained. Consequently, endocrine disrupting mechanism of 2,4-D and Atrazine was concluded.
