叶下珠药材指纹图谱的研究

【摘要】 叶下珠药材指纹图谱的研究 中药指纹图谱是指运用现代分析技术对中药化学信息以图形(图像)的方式进行表征并加以描述。通过反映中药内在化学成分的种类和数量,提供全面的、综合的信息以控制中药材及中药制剂的质量,是目前国际上能认可的、较好的反映了中药材及中药制剂质量的质量控制方法。 叶下珠(又名珍珠草),为大戟科植物叶下珠(Phyllanthus urinaria L.)的全草,民间常用于治疗腹泻、下痢、尿路感染、肾炎水肿和黄疸性肝炎等疾病,近年来国内外均将其用于治疗病毒性乙型肝炎,被认为是一种有广阔发展潜力的安全有效的抗乙肝病毒植物药。本文以叶下珠药材中所含的酚酸类成分柯里拉京和没食子酸为对照品,针对其中的水溶性成分和脂溶性成分采用不同的提取方法,考察了甲醇—水流动相系统和乙腈—水流动相系统,并通过加入小分子有机酸(如乙酸、三氯乙酸)以及四氢呋喃等优化流动相系统,以改善色谱峰的峰形和分离度,通过反复摸索确定了流动相洗脱梯度,使药材中的水溶性和脂溶性成分在取得良好分离度的基础上保持适当峰数和稳定的色谱峰峰面积积分值,经过制备、分析、比较、评价和校验过程,建立了叶下珠药材的高效液相色谱指纹图谱和标准,同时比较了不同产地出产的叶下珠药材之间的差异。结果表明,叶下珠药材指纹图谱能够反映药材之间的相似性,为叶下珠药材的质量控制提供了可靠依据。同时,通过比较不同产地的叶下珠药材的指纹图谱之间的差异,可以看出不同产地的叶下珠药材之间的差异较小。造成差异的原因可能是药材生长环境,采收季节、贮藏时间的长短不同,以及取样均匀性、提取条件不能保持完全一致。
【Abstract】 The fingerprint reserch of medical material of Phyllanthus urinaria L.The modern analytical technique is applied to the reserch of fingerprint by using figures to describe the internal chemical information of the traditional Chinese medicine. The fingerprint can reflect the different of description and quantity of chemical information. It can be used to control the quality of the traditional Chinese medicine and drugs preparati on through the full-scal e and integrative information. It’ s a better method to control the quality of traditional Chinese medicine which could be ratified in the international.In Chinese civil, Phyllanthus urinaria L. is used to treat the coprorrhea, alv laxat, urinary tract infection, oedema nephriticum, icterohepatitis, and so on. In the past few years , it is mainly used to cure the Type B viral hepatitis and known as a potential, safe and effective plant amedica. In this articles, we used two of the internal phenolic acid ingredient-cotilagin and gallic acid-as control article. We used different extract methods and considerated two mobile phase systems-MeOH-H2O and acetonitrile-H2O.For the purpose of optimize the mobile phase systems, we added micromolecule organic acid, such as acetic acid, trichloracetic acid and tetrahydrofuran to improve the peak shape and resolution. We ascertained the the mobile phase systems and elution grad. The water-solubility and liposolubi1ity ingredient were maintained proper resolution under the circumstance. All chromatographic peak area score are stable. Through the process of evelopment analysis, comparision evaluation,andverification,we established the HPLC fingerprint of Phyllanthus urinaria L. and compared Phyllanthus urinaria L. which yielded from diferent places. The consequence indicate that the fingerprint can reflect the resemblance of the medical material and provide re I iable evidence to control the quality of Phyllanthus urinaria L. At the same time ,we compared the distinction of fingerprint of Phyl lanthus urinaria L. which yielded from different places and found thatthere are no significant difference. These minor difference may he caused by the different living surrounding, collected season, the period of provision, and the homogeneity of taking of samples and the condition of extraction which we can’t hold completely at equal pace.
