低能离子注入拟南芥的诱变效应和机理研究

【摘要】 以拟南芥为对象做离子注入的物理诱变,观测记载了离子束注入后供试材料的形态性状变化,利用RAPD方法探讨了引起形态性状诱变的分子机理,结合同工酶分析结果,分析了离子束辐照引起的生理诱变效应。 研究发现,离子束辐照拟南芥干种子引起了多方位的诱变效应。 1.表现在M1代发芽率的影响。发芽率-剂量曲线是“马鞍型”曲线。另外,M1代营养器官(株高、叶长、叶宽)的数量指标损伤度随着剂量的变化而不同,最显著地表现为株高明显降低。高剂量仅为对照的55%左右。 2.通过扫描电镜对离子注入后的拟南芥种子的观察发现,经离子注入过的种子种皮有明显的孔洞和裂痕,并且注入离子对干种子表皮的作用程度随剂量的增大而加深。 3.通过对离子注入后拟南芥过氧化物同工酶活性的测定和酶谱的分析表明,其活性的大小、酶谱谱带的有无及深浅与剂量的关系和苗期性状指标与剂量的关系是一致的。 4.利用RAPD方法揭示了离子注入对拟南芥DNA多态性的影响,并在分子水平上揭示了辐照的不同剂量对拟南芥遗传多态性的影响,阐明了离子诱变的分子机制。结果显示,剂量组所扩增的差异条带率随着剂量的增加而增加。

【Abstract】 By using Arabidopsis thaliana as material, the study had been focused on the ion beam implantation -induced effects and the changes of the configuration. With the RAPD method and the analysis of the POD, the mechanism in molecular biology and physiology were stated.The ion beam implantation induced aberration of Ml in many aspects. Firstly, it was displayed dominantly in the germination rates. The germination rate-dose function curve is down in the first, then up and down at last. Moreover, the injury rates of the nutrient organs (the height of the plants, the lengthen and the width of the leaves) were different when the dose was changed. One of the most evident aspects is the short of the plant lengthen, in the big dose was only 30 percent of the CK. Secondly, the observation of the Arabidopsis thaliana seeds which were irradiated by the ion beam through the SEM found that with the increase of the ion beam dose, the ion beam mechanical erosion degree of the seeds was deep. And there are many holes and gaps in the surface of the seeds in dose. Thirdly, the changes of peroxidase(POD) activity and the analysis of the enzyme spectrum in the morphological different stage in the Arabidopsis thaliana were tested. The result indicated that the relation of dose and the activity was consistent with that of dose and the injury rates of the nutrient organs. Lastly, the RAPD method make the effects of the ion beam irradiation induced the Arabidopsis thaliana DNA clear. And in the molecule level explain the results of the different doses act on the Arabidopsis thaliana DNA and the molecule mechanisms.
