魔芋葡甘聚糖酶产酶菌的选育及所产酶性质的研究

【摘要】 随着生活水平和质量的提高,人们开始越来越重视自身的健康和保健问题。许多具保健功能的自然资源,日益为人们所关注。如何开发?尤其对像魔芋这样适宜栽植于山坡地、资源丰富的作物?开发高附加值产品是其关键。魔芋低聚糖现已成为人们关注的焦点,其生产无论从经济还是现实的角度来看,酶法降解已成为大势所趋,过去在自然界中筛选培育的菌株,酶活性往往太低,不利于工业化生产。因此,能否筛选出高产优质菌株已成为该产业链中的制约瓶颈。 【目的】以自然界中得到的菌株为出发株,诱变选育出高产葡苷聚糖酶的菌种,要求菌株须生长快,发酵周期短,不易为杂菌污染,为低聚糖的产业化生产提供技术支撑和经济保障。 【方法】应用常规细菌培养、分离、生理与生化鉴定方法分离目的菌株;应用生化法测定目的菌株的产酶活力并对所产酶的性质进行了分析。 【结果】 1、发现Brachybacterium sp A6所产酶对葡甘聚糖酶解速率快。 2、发现pH值7.0,碳源2.5%,培养温度36℃时为Brachybacterium sp A6最适培养条件。 3、发现酶活性及稳定性可随培养条件的改变而发生变化。 【结论】 1、以从自然界中筛选出的菌株为出发株,经诱变、筛选,得一高产葡甘聚糖酶菌株,初步鉴定为短杆菌属Brachybacterium sp A6. 2、经诱变、三角瓶培养,该菌株的最适培养条件:培养基pH值7.0,碳源2.5%,振荡培养,200r/min,培养温度36℃,培养48h。发酵液酶活力达340u/ml,高于以往文献报道水平。 3、酶活性及稳定性随环境而变化,在pH6.0和温度55℃时,酶的活性最高,温度在0-50℃之间变化时,温度越低,稳定性越大。pH值在5-8间变化时,pH值 越大,稳定性越好。溶液的离子强度、种类对酶活性有很大的影响,适当浓 度的 MnD门.l刁)对此粗酶有激活作用,Fek、AIl、EDTA、Mg卜对粗酶则 有强烈抑制作用。

【Abstract】 With the development of people’s live standard,much importance are attached to health food,how to explore these natural source,particularly of Amorphailsu.the key lies in finding high valued production,in this aspect,how to get high activity enzyme is the prerequisite.ObjectionSelecting,inducing demanded strain (high enzyme productive),originated from nature,short ferment cycle,high resistance to environment factors.Measuresbacteria culturing,separating,confirming,analysis of activity and properties of Enzyme-ManrianseResultsBrachybacterium sp A6 is high productive in generating Enzyme-Mannanse,particularly in Amorphailsu.pH7.0,temperature 36 C,Carbon content 2.5%,suitable for Brachybacterium sp A6.Activity and stability of Enzyme-Mannanse changed according to different cultural condition.ConclusionA systematic method for preparation of Enzyme-Mannanse is established,a high productive strain was got after seducing and selecting from nature,confirmed as Brachybacterium spA6Research were conducted on medium and culture method of the strain in order to get the suitable cultural condition of fermentation,the experiment result shows the optimium condition is pH7.0,temperature 36C;Carbon content 2.5%,ventilation in abundence,agitation speed 200r/min. enzyme activity of Mannanse reached 340u/ml (250ml),a leader in today’s china.Activity and stability of Enzyme-Mannanse changed according to variety of cultural condition. The optimum condition of enzymes activity is in pH6.0,temperature 55 C .Enzyme activity also changed according to different kind of Ion and Ion content,Mn2+ positive eflfect;Fe3+,Al3+,EDTA,Mg2+ negative effect.
