甘氨酸螯合锌食品添加剂的研究

【摘要】 锌是人体必需的微量元素,锌以蛋白质和金属酶的形式存在于人体中,履行着各种重要的生物功能。膳食中锌的主要来源为动物性食品,而我国传统的饮食结构是以植物性食品为主,加上人们营养保健观念的淡薄很容易造成缺锌而导致各种病理变化和疾病的发生。针对这种状况,在膳食外的适量补锌是非常必要的。甘氨酸锌符合人体锌吸收的机制和特点,把人体必需的甘氨酸和微量元素锌有机的结合起来,成为一种新型高效的锌营养强化剂。本课题以甘氨酸锌和氢氧化锌为原料通过化学方法合成甘氨酸整合锌,产品经纯化后测定其部分基本理化性质常数,并将其应用到豆油中进行抗氧化试验。 化学方法合成甘氨酸螯合锌以pH值、温度、配位比、浓度为反应的主要影响因素,以产率为评价指标,经正交试验确定最优反应条件为:pH=8.5,温度为60℃,配体摩尔比为Gly:Zn=2:1,甘氨酸浓度为15%。该反应过程制备的甘氨酸螯合锌产率为52.6%。产品经纯化后,分别测定了其元素分析和紫外一可见、红外光谱学性质,初步确定产品即为甘氨酸螯合锌。再经过影像板单晶X射线结构分析仪测定其晶体结构参数:分子式为:C4H10N2O5Zn;结构式为:[Zn(H2NCH2COO)2]2·H2O;相对分子质量:M=231.51,晶系属于单斜晶系,空间群属于C2/C群,晶胞参数:a=19.716(3)A,b=10.4590(11)A,c=15.0965(18)A,alpha=90deg.,beta=92.998(3),gamma=90deg.,体积V=3108.8(7)A3,F(000)=1888,Z=16,密度Dc=1.979g/m3。 甘氨酸螯合锌的熔点为:282~284℃。甘氨酸螯合锌在37℃时的溶解度为41.35mg/100g水。在室温下,pH=8.5时,利用分光光度法测得甘氨酸锌水溶液的稳定常数为5.54。 按0.02%的添加量分别向油脂中加入甘氨酸锌和BHT,在90℃储藏条件下空白、BHT、甘氨酸锌达到诱导期的时间依次为9.2、10.7、11.3h,初步表明甘氨酸锌在强化豆油锌的同时还具有抗氧化作用。

【Abstract】 Zinc is the microelement necessary for human body, which exists in human body in the manner of protein and metal enzyme, performing various important biological functions. The zinc carrent in human body directly control various process of metaboly, if there is disorder in the process of metaboly in zinc ingestiom absortiom storage, using and excretion etc, it will lead to various pomology change even disease. Microelement is not like other things, which can not make in human body but only can acquire from outside body. Only a little zinc can be absorbed, food zinc can be absorbed by body are less than 10%.So, we must supply zinc reasonably. Zinc-glycine chelate is a new nutritional intensifying agent which has higher bioavailability than inorganic zinc.The zinc-glycine chelate was synthesized with glycine and zinc hydroxide as raw materials.The optimum technological conditions are that molar ratio of glycine to zinc hydroxide was 2:l,reac-tion temperature was 60C,reaction pH was 8.5,and reaction time was 2h.The yield was 52.6%.Composition and structure of the complex were characterrrized by elementary analysis,infrared spectrum,thermogravimetric and X-ray powder diffractionJts imposition is [Zn(NH2CH2COO)2] H2O. The results of indexization cf the X-ray powder diffraction data indicated that the crystal structure of the complex belongs to monoclinic system. The lattice parameters are: a=19.716(3)A, b=10.4590(ll)A,c=15.0965(18) A, alpha=90deg., beta=92.998 (3), garnma=90deg., V=3108.8 (7) A3, Z=16,DC=1.979g/cm3.The constants of zinc-glycine chelate were measured.The solubility of zinc-glycine chelate is 41.35mg/100gH2O.The melting point of zinc- glycine chelate is 282-284 C.The logarithms of stability constant in aqueous solution atpH=8.5 is 5.54. The oxidation resistance of zinc-glycine chelate was tested in soybeanoil.The result indicates that zinc-glycine chelate has higher ability of oxidation resistance than BHT in soybean oil.
