【摘要】 目的： 1．从细胞凋亡角度探讨六神丸治疗白血病的机理，为临床应用提供依据。 2．老药新用，扩大六神丸的应用范围。 方法： 1．中药血清药理学方法制备六神丸含药血清。 2．以乙醇为溶媒，沙氏回流法提取六神丸。 3．MTT法测定六神丸对HL-60细胞的增殖抑制作用。 4．吉姆萨染色法观察细胞凋亡的形态学变化。 5．原位末端标记（POD法）检测细胞凋亡。 结果： 1．MTT法表明：六神丸乙醇提取剂对HL-60细胞有明显的增殖抑制作用，且呈浓度依赖关系，IC₅₀为0.5μg／ml；中剂量组所得六神丸含药血清对HL-60细胞增殖抑制优于低剂量组和高剂量组。 2．吉姆萨染色显示六神丸乙醇提取剂和六神丸含药血清处理过的HL-60细胞均出现典型的凋亡形态，如细胞膜皱缩、染色质固缩碎裂、凋亡小体形成等。 3．原位末端标记表明：六神丸乙醇提取剂诱导HL-60细胞凋亡在一定范围内呈时间-浓度依赖关系；六神丸含药血清诱导HL-60细胞凋亡作用优于As₂O₃血清组和空白血清组（P＜0.01）。 结论： 六神丸体外可以诱导HL-60细胞凋亡，诱导细胞凋亡是六神丸治疗白血病的可能机理之一。更多还原

【Abstract】 Purposel.To explore the mechanism of liu-shen-wan in treating AML from inducing apoptosis and to supply the theorical basis in clinical therapy.2.To enlarge the clinical application of liu-shen-wan. MethodI.Chinese herbs serum pharmacology method produces medicine serum of Liu-Shen-Wan2.Sha shi distillation method extracts Liu-Shen-Wan, with alcohol as the solvent.3.MTT assay detects the growth inhibition effect of Liu-Shen-Wan on HL-60 cells.4.GIMSA dying assay observes morphology changes of apoptosis.5.In situ end-labeling method (POD) probes apoptosis phenomena Resultsl.MTT assay indicates:Liu-Shen-Wan alcohol extraction has obvious growth inhibition effect on HL-60 cells, and has dependency on concentration. ICso is 0.5ug/ml. As far as the growth inhibition effect is concerned, the middle-dose group of Liu-Shen-Wan medicine-serum is superior to the low-dose group and high-dose group.2.GIMSA dying assay exhibits: HI-60 cells dealed with Liu-Shen-wan alcohol extraction and Liu-Shen-Wan medicine-serum presents typical apoptosis morphology, such as cell membrane shrinkage, chromatin contraction and rupture, apoptosis body formation.3.In situ end-labeling method (POD) indicates:HL-60 cells apoptosis induced by Liu-Shen-Wan alcohol extraction have time-concentration dependency relation within certain ranges; The HL-60 cells apoptosis effect induced by Liu-Shen-Wan medicine-serum is superior to AS2O3 serum group and blank serum group.(P<0.01)ConclusionsLin-shen-wan can induce apoptosis in HL-60 cells in vito.Inducing apoptosis is one of possible mechanisms in treating AL by Liu-Shen-Wan.更多还原
