纤维素酶解饲料酶源菌筛选及酶作用特性的研究

【摘要】 本研究从青海互助北山森林土样中分离筛选到产纤维素酶真菌No.0143菌株,根据其形态学特征鉴定,定种为康氏木霉(Trichoderma koningii Qudem,T.koningii),其湿固体发酵物含15u/g滤纸酶活力(FPA)。No.0143菌株经亚硝基胍诱变后,获得一株突变株No.0143p,其湿固体发酵FPA为20u/g,是出发菌株的1.33倍。再对No.0143p菌株进行产酶条件优化及酶作用特性的研究,试验结果表明:(1)T.koningii No.0143p菌株在麦麸为碳源、尿素为氮源、起始pH 7.0、28℃培养6天的条件下产酶最高,以此产酶优化条件处理No.0143p菌株,其湿固体发酵物FPA可达32u/g,为出发菌株的1.6倍;(2)酶的最适作用温度和pH分别为50~55℃和4.8。综上可见,T.koningii No.0143p菌株可作为纤维素酶解饲料酶源菌,具有良好的饲用前景,可做进一步的酶化纤维素饲料研究。

【Abstract】 A strain Trichoderma koningii No.0143 which produces cellulase was isolated from a soil sample of the east area in QingHai, its FPA activity was 15u/g. After treatment No.0143 strain with NTG, a mutant (No.0143p) was obtained whose FPA activity was improved from 15u/g to 20u/g. After selected carbon and nitrogen resources, the incubated time, pH and temperature, the FPA activity was improved further to 32 FPA units. It was proved that wheat bran, urea, pH 7.0, 28℃It was showed that No.0143p strain was promising fungi in enzymatic feed. The cellulase optimum PH and temperature were 4.8. and 55℃ respectively.
