以毛泡桐为原料提取、分离纯化熊果酸的工艺研究

【摘要】 熊果酸属于有机三萜酸,具有镇静、抗炎、增强机体免疫力、抗菌、美白、抗癌等药理作用,近年更被中国医学科学院肿瘤医院实验证明其有望成为低毒、有效的新型天然抗癌药物,因而正日益受到众多学者的重视。 本课题为以毛泡桐为原料提取、分离纯化熊果酸的工艺研究,详细总结了熊果酸的原植物研究近况、熊果酸的药理作用研究进展以及其分离纯化的方法;摸索出从毛泡桐叶、柄、花中提取、分离纯化熊果酸的工艺流程和最佳工艺条件;分析了不同采集期、不同部位毛泡桐中熊果酸含量的变化规律,为原料的采收提供依据;产品经中国医学科学院药物研究所分析熊果酸含量为96.9%。 所得工艺条件为: (1)毛泡桐叶晒干后粉碎,12倍量无水乙醇回流提取10小时,减压回收溶剂得浸膏,15~20倍量石油醚除杂3~4次,每次20小时,乙醇精制3-4次; (2)硅胶柱色谱甲醇—水(8:1)梯度洗脱,无水乙醇重结晶; (3)HPLC法分析不同采集期毛泡桐叶中熊果酸含量,结果以9月含量最高,7月次之,5月最低。

【Abstract】 Ursolic acid belongs to triterpenic acid, it has tranquihzation, anti-inflammation, strength immunity , anti-bacteria, beautiful white and anti-tumor pharmacological actions. Thus much more attention has been paid to the studies of Ursolic acid since it was experimental proved having the hope to become a new potent natural anti-cancer medicine of low-poison & high-validity by the tumor hospital of Chinese acadmy of medical in recent decades.This paper is studying on the process of isolating & purifying Ursolic acid from P.tomentosa(Thunb.)Steud. It detailedly summarize the current research situation of the original plants > development in researches of Ursolic acid in pharmacological actions and the methods of separation and purification. A practical process ilow and the optimal process of extracting Ursolic acid from P.tomentosa(Thunb.)Steud. leaves -, stems & flowers were afforded by doing lots of experment. Analysing the change regulation of Ursolic acid content in samples gathered in different periods and parts, which offer the basis for collection. The content of Ursolic acid in the product obtained was determined by HPLC, the: result is 96.9%. The process obtained is:(1) Dried powder of P.tomentosa (Thunb.)Steud. leaves were extracted with 12 times absolute EtOH for 10 hours and the EtOH solution was concentrated under reduced pressure to give a crude residue which was suspended in 15-20 times PE (Bp 60-90 ) for 3-4 times (20 hours for each times) and then purified by EtOH for 3-4 times.(2)The residue obtained was subjected to column chromatography on silica gel and eluted with MeOH/H2O gradient system (8:1) followed by recrystallization in absolute EtOH finally afforded a colorless needles.(3) The content of Ursolic acid in P.tomentosa(Thunb.)Steud. gathered in different periods was determined by HPLC the result shows that the content appears to be highest in samples gathered in September lower in July and lowest in May.
