不同剂量小檗碱对大鼠缺血心肌的保护作用

【摘要】 目的验证小檗碱是否对大鼠梗死后心脏具有保护作用,并初步探讨小檗碱的这种保护作用是否随剂量的改变而有统计学差异。方法将56只大鼠随机分为7组,每组各8只大鼠。即:假手术组,模型组,小檗碱100mg/kg,小檗碱150mg/kg,小檗碱200mg/kg,小檗碱250mg/kg和普奈洛尔组。各组大鼠均按照自己组规定药品剂量进行灌胃,总共灌胃7天,每天灌胃1次。每组大鼠均于第2天灌胃4小时后,采用开胸结扎冠脉方法制作心肌梗死模型。模型建立成功后,各组大鼠继续灌胃5天。每组大鼠均于第7次灌胃后4小时测量相关心功能参数,之后收集心脏和血液标本作相关项目分析。结果小檗碱可以显著改善大鼠梗死后的心脏功能,显著减小大鼠梗死后心脏的缺血范围和梗死范围并显著降低大鼠血清中心肌乳酸脱氢酶(LDH, lactate dehydrogenase)的含量。且在实验条件下,不同剂量小檗碱对大鼠梗死后心脏的保护作用有统计学差异。结论小檗碱对大鼠梗死后心脏有显著的保护作用,且小檗碱的这种保护作用随剂量的不同有统计学差异。
【Abstract】 Objective:To testify berberine (Ber) has or not the protective effect on the infacted heart on the rat. And to explor initially the protective effect of berberine with the different doses have the different results.Methods:56Wistar rats were randomly divided into7groups, there were8rats in each group. There are the sham operation group, the model group, the berberine with100mg/kg group, the berberine with150mg/kg group, the berberine with200mg/kg group, the berberine with250mg/kg group and the Propranolol group. The rats were given drugs by the gavage method with different medicines and different dosees. All rats would be gavaged7days. And the frequency was one time a day. Using the thoracotomy and the coronary artery ligation to establish the myocardial infarction models on the second day after the first gavage. All rats were killed after measuring the relevant parameters of the heart functions. And the hearts and blood samples were collected for the analysis.Result:Berberine could improve obviously the function of the rats, and reduce the infarct and the ischemia sizes in the hearts of the rats. And it also decrease the level of LDH in the blood of the rats. And under the experimental conditions, the results of the groups of berberine different doses had significant difference.Conclusion:Berberine can significantly improve the function of the infarction heart in rats, and the function of berberine with different doses have significant difference.
