繖序臭黄荆及海星(ZDLDWO32)化学成分研究

【摘要】 本论文分为繖序臭黄荆化学成分的研究和海星(ZDLDW032)化学成分的研究两部分。利用硅胶柱色谱(CC)、薄层色谱(PTLC)、反相柱色谱(ODS)、葡聚糖凝胶色谱(Sephadex LH-20)和高效液相色谱法(HPLC)等现代分离技术,从这两种常用中药材中得到了23个化合物。利用核磁共振(NMR)、红外光谱(IR)和质谱(MS)等现代波谱技术,确定了19个化合物的结构。繖序臭黄荆(Premna serratifolia L)属马鞭草科(Verbenaceae)植物,民间常用来治疗蛇咬伤,创伤出血,腰腿痛,跌打损伤,牙痛,烧伤及疟疾,痢疾等症。从繖序臭黄荆中分离了18个化合物,鉴定出其中15个化合物的结构,分别是:piscrocins D(1)、rehmaglutinA(2)、10-O-(Z)-p-coumaroylcataipolD(3)、10-O-(E)-p-coumar-oylcataipo(l4)、β-蜕皮激素(5)、Integrister-one A 20,22-acetonide(6)、β-谷甾醇(7)、β-胡萝卜甙(8)、芫花素-5-O-β-D-葡萄糖苷(9)、槲皮素(10)、山奈酚-3-O-β-D-半乳糖苷(11)、芹菜素(12)、3′,5,7-三羟基-4-甲氧基黄酮(13)2-Hexylidene-3-methylsuccinic acid(14)、2′-脱氧胸腺嘧啶核苷(15),化合物1~15均首次从繖序臭黄荆中分离得到。海星学名海盘车,属棘皮动物门(Echinodermata)海星纲(Asteroidea),为海洋常见大型无脊椎动物之一,是研究海洋药物的良好材料。海星的活性成分及其作用机理的研究吸引了越来越多的学者,目前已从海星中发现多种类型的化合物,其中有蛋白质、酶、甾类糖苷、神经节苷酯、生物碱等生物活性物质,这些化合物具有抗癌、抗肿瘤、抑菌、鱼毒和抗氧化等活性。从海星(ZDLDW032)中分离了5个化合物,鉴定了其中4个化合物的结构,分别是:amurensoside D(16)、胆固醇(17)、24-亚甲基胆甾醇(18)、1-十八烷基甘油醚(19)。

【Abstract】 Two parts are involved in this dissertation : The studies on the chemical constitu- ents of Premna serratifolia L and The studies on the chemical constituents of Starfish (ZDLDW032). These two commonly used Chinese herbal medicines were isolated by modern chromatographic methods including CC, PTLC, ODS, Sephadex LH-20 and HPLC, and twenty-three compounds were gained. The structures of Nineteen compounds were elucidated by spectroscopic methods including NMR , IR and MS.Premna serratifolia L, which belongs to the family of Verbenaceae, commonly used to treat people bitten by snakes, post-traumatic bleeding, pain, bruises, toothache, burn , malaria, dysentery etc. Eighteen compounds were isolated from the Premna serratifolia L and Fifteen compounds in them were identified as:piscrocins D(1), rehmaglutinA(2),10-O-(Z)-p-coumaroylcataipol(3), 10-O-(Z)-p-coumaroylcataipol(4),β-ecdysone(5), Integrister-oneA20,22-acetonide(6),β-sitosterol(7),β-daucosterol(8), Genkwanin-5-O-β-D-glucopyranoside(9),Quercetin(10),Kaempferol-3-O-β-D-galact-opyranoside(11),Apigenin(12),3′,5,7-trihydroxy-4-methoxylflavonoid(13),2-hexylidene-3-methyls-ccinic acid(14), thymine-2-desoxyriboside(15).Starfish (phylum Echinodermata, class Asteroidea), which is one of the common large invertebrate, have proven to be an especially valuable source. The investigation of active component and mechanism of action of Starfish draws more and more scholars. At present, many polytype compounds have already been discovered. Starfish contains different types of bioactive materials, such as proteins enzymes, steroidal glycosides, gangliosides, alkaloids etc. These compounds have activity of anticancer, antioncoma, bacterio-stasis, ichthyotoxin, antioxygen and so on. Six compounds were isolated from the Starfish(ZDLDW032) and Five compounds were identified as:amu- rensoside D(16), cholesterol(17), Ergosta-5,24(28)-dien-3β-ol (18), batyl alcohol(19).
