54个广西区试甘蔗品种的抗旱性评价及甘蔗抗旱基因的分离

54 Guangxi Regional Trial Sugarcane Varieties of the Drought and Drought Resistance Gene Analysis of the Separation

【摘要】 本研究主要有两部分内容:1.以进入区试的54个甘蔗品种作为试验材料,在苗期进行自然干旱胁迫,在甘蔗表现出重度干旱情况下,测定甘蔗正1叶的叶绿素相对含量、叶片相对含水量、超氧化物歧化酶(SOD)活性、叶片水势、丙二醛(MDA)含量、游离脯氨酸(Pro)含量、过氧化物酶(POD)活性等生理生化指标。以复水恢复情况为参考,采用隶属函数和主成分综合分析方法,对供试54个甘蔗品种的抗旱性进行综合评价。结果把54个甘蔗品种苗期抗旱性划分为三类(高度抗旱、中度抗旱、不抗旱):(1) 13个高度抗旱品种:GT467、GT1247、GT02-235、ROC22、GT496、粤糖57-423、G28、NCO310、GT1156、GT649、ROC10、GT97-69、GT2357。(2)20个中度抗旱品种:GT3、GT21、GT99-107、GT02-761、GT2625、GT02-899、赣蔗18号、园林17号、GT208、GT97-40、GT963、GT342、GT1237、GT02-770、柳03-1137、B1、GT02-833、F172、G17、B8。(3)21个不抗旱品种:粤糖93-159、GT02-281、园林6号、B28、GT02-413、GT02-901、GT02-351、柳04-12、GT123、GT11、ROC16、GT133、柳03-182、GT99-181、柳03-255、GT619、GT02-237、GT390、GT02-469、GT1403、GT02-99。2.选取苗期高抗旱品种ROC22和低抗品种ROC16为材料,在伸长期进行干旱胁迫处理,提取叶片总RNA后,经反转录合成第一链cDNA,采用SRAP (Sequence-related amplified polymorphism相关序列扩增多态性)标记,用108对SRAP引物组合进行甘蔗抗旱差异基因筛选,结果表明:(1)在甘蔗上建立了cDNA-SRAP标记的最佳的PCR扩增反应体系、退火温度及反应程序,为甘蔗SRAP标记分离更多的抗旱基因奠定了基础。(2)用108对SRAP引物组合对苗期ROC22和ROC16两品种进行抗旱性筛选,得到11个差异片段;经再扩增、纯化和测序,得到1条吡咯啉-5-羧酸合成酶抗旱基因的差异片段。

【Abstract】 This research was divided into two parts:Firstly,54 sugarcane varieties were studied at the seeding stage under natural drought stress in order to explore the changes of the physiology and biochemistry indexes of the +1 leaves like relative content of chlorophyll, relative water content,leaf water potential, SOD/POD activity, the content of MDA or pro and so on. The membership function and principal constituent analysis methods were adopted to estimate the ability of drought resistance of the 54 sugarcane varieties under the water recovery condition. And the result showed that they were divided into three groups:highly drought resistance, moderate drought resistance and the disability of drought resistance.There are 13 highly drought resistance varieties, GT467、GT1247、GT02-235、ROC22、GT496、YT57-423、G28、NCO310、GT1156、GT649、ROC10、GT97-69、GT2357.20 moderate drought resistance varieties are:GT3、GT21、GT99-107、GT02-761、GT2625、GT02-899、GZ18、YL17、GT208、GT97-40、GT963、GT342、GT1237、GT02-770、L03-1137、B1、GT02-833、F172、G17、B8。And the disability drought resistance varieties are:YT93-159、GT02-281、YL6、B28、GT02-413、GT02-901、GT02-351、L04-12、GT123、GT11、ROC16、GT133、L03-182、GT99-181、L03-255、GT619、GT02-237、GT390、GT02-469、GT1403、GT02-99.Secondly, RNA was extracted from the highly drought resistance variety ROC22 and the disability drought resistance variety ROC 16 which were treated under water stress at the elongation stage. And reverse transcription was carried out to obtain cDNA template, then adopted the Sequence-related amplified polymorphism (SRAP) marker to sift out the drought resistance gene with 108 pairs of SRAP primers. And the result showed that:The best amplification system and conditions of sugarcane SRAP mark were set up which were do a great favor to separate more drought resistance.11 differential sequences were sifted from the ROC22 and ROC 16 with the 108 pairs of SRAP primers. And one Pyrroline-5-carboxylate dehydrogenase differential sequence was obtained after re-amplification, purification and sequencing.
