共同性外斜视内直肌IGF-Ⅰ、AnnexinA1的表达研究

【摘要】 目的观察共同性外斜视眼外肌中IGF-1、Annexin A1表达的变化,探讨IGF-1、Annexin A1在共同性外斜视发病机制中的作用。方法收集我院2009-2010年收治的共同性外斜视病人(16例)为斜视组。根据年龄又将斜视组分为儿童斜视组(8例)和成人斜视组(8例);同期行眼球摘除或角膜移植供体取得的水平直肌为成人对照组(共6例)。所有眼外肌固定切片后行IGF-1、Annexin A1免疫组织化学染色,测IGF-1、Annexin A1在眼外肌中的平均光密度值。对各组间平均光密度值进行比较,并对斜视患者眼外肌中IGF-1平均光密度值与患者斜视度及年龄进行相关性分析。结果①IGF-1在斜视组、成人对照组、儿童斜视组及成人斜视组表达的平均光密度值分别为(0.225±0.041)、(0.334±0.024)、(0.255±0.012)及(0.196±0.038)。斜视组与成人对照组、儿童斜视组与成人斜视组、成人斜视组与成人对照组之间比较均有统计学意义(P<0.05)。②Annexin A1平均光密度值在斜视组、成人对照组、儿童斜视组及成人斜视组表达的平均光密度值分别为(0.148±0.031)、(0.254±0.050)、(0.197±0.027)及(0.126±0.026)。斜视组与成人对照组、儿童斜视组与成人斜视组、成人斜视组与成人对照组之间比较同样均有统计学意义(P<0.05)。③斜视患者IGF-1、AnnexinAl平均光密度值与患者的斜视度和年龄呈负相关(r=-0.721,P=0.002,r=-0.663,P=0.008;r=-0.554,P=0.029,r=-0.598,P=0.014)。结论①眼外肌中IGF-1、Annexin A1表达的降低可能参与斜视的发病。②眼外肌中IGF-1、Annexin A1中的表达变化可能与斜视的发展相关。

【Abstract】 Objective To investigate the expression of IGF-1 and Annexin 1 of extra ocular muscles (EOMs) in patients with concomitant extropia. To explore the role of IGF-1 and Annexin lin the etiology of concomitant extropia.Methods Collect 16 cases accepted surgery with concomitant extropia and name strabismus group, then divided them into 2 groups which were the group of children with concomitant extropia and the group of adults with concomitant extropia. Medial rectus muscles were obtained with agreement from patients accepted surgical resection. Control muscles were obtained from patients with eyeballs removed with injury and corneal transplant donors. Cross sections were stained immunochemically and measured the mean optical density according to the expression of IGF-1 and Annexin 1 in EOMs. To compare the difference in different groups and analyze the relationship between the expression of IGF-1 and Annexin 1 and the age and the strabismus degree.Results①We discovered IGF-1 and Annexin 1 expression in EOMs of strabismus group and control group. There were significant differences (P< 0.01) in the expression of IGF-1 and Annexin A1between the group of children with concomitant extropia and the group of adults with concomitant extropia, the control group and the group of adults with concomitant extropia, the group of adults with concomitant extropia and the strabismus group. The formers had higher level than the latters.②There were significant differences (P< 0.01) in the expression of IGF-1 and Annexin A1 between the strabismus group and the control group. And the former was lower than the latter.③There were negative relationship between IGF-1 and Annexin A1expression and prism diopter of deviation and patients’ age.Conclusion The decreased level of IGF-1 and Annexin 1 expression may participate in the incidence of concomitant extropia by inhibit the mechanism of self-renewal and repair; With age, the level of IGF-land Annexin A1 is down gradually, we consider it may be the cause of concomitant extropia with deteriorating process.
