规模化鸡场消毒剂的筛选与消毒效果测定

【摘要】 规模化鸡场疫病的发生往往是多因素综合作用的结果,但其中最主要的是由于外界环境中病原微生物的侵入及扩散或场内鸡群本身病原微生物污染扩散造成的。通过测定消毒剂A(5%苯扎溴铵)、消毒剂B(2%戊二醛溶液)、消毒剂C(10%聚维酮碘)、消毒剂D(10%癸甲溴铵)和消毒剂E(过硫酸氢钾复合物粉)5种消毒剂的杀菌能力,筛选出杀菌能力强的消毒剂,随后对筛选出的消毒剂进行消毒效果测定,为规模化鸡场正确应用消毒剂进行疫病预防和制定消毒程序提供参考。采用有限稀释法测定5种消毒剂对参考菌株(大肠杆菌8099)和鸡舍中分离的大肠杆菌的最小杀菌浓度(MBC);菌悬混合液定量杀菌试验测定5种消毒剂对参考菌株的杀灭效果;采用自然沉降法测定鸡舍空气中细菌含量,经鸡舍带鸡消毒试验,综合测定消毒剂的消毒效果。试验结果表明,过硫酸氢钾复合物粉的MBC值最小,杀菌作用最强,癸甲溴铵次之,聚维酮碘、戊二醛、苯扎溴铵杀菌作用较弱;5种消毒剂与菌悬液分别作用1 min、5 min、10 min,消毒剂E对大肠杆菌的杀灭效果均极显著优于聚维酮碘、戊二醛、苯扎溴铵(P﹤0.01),但与癸甲溴铵对大肠杆菌的杀灭效果差异不显著(P﹥0.05)。鸡舍现场消毒试验表明,用过硫酸氢钾复合物粉对鸡舍中空气、墙壁、地面、饲槽、饮水器以及鸡笼表面具有显著的消毒效果,消毒对鸡死亡率的影响差异显著(P﹤0.05),消毒后试验组鸡的死亡率比对照鸡群平均下降了2.86%。说明消毒剂E消毒效果好,适合于规模化鸡场带鸡喷雾消毒。

【Abstract】 The epidemic of scale chicken farm is the results of comprehensive factors caused, but the main factor is the invasion and diffusion pathogenic microorganism from external environment or pathogenic microorganism diffusion in chicken themselves. Disinfection potential of five disinfectants (A, B, C, D, E) were determined for screening the disinfectants with strong disinfection potential, and then the efficacy eveluation of disinfection was made to these screened disinfectants. Based on above results, it will provide reference for chicken house to use correctly disinfectants for epidemic prevention and establishing disinfection procedure.The minimum bactericidal concentration(MBC) of five disinfectants to reference strains (Escherichia Coli 8099) and E.coli isolated from chicken house was determined by limiting dilution assay. The result of five disinfectants to antisepticize reference strains was determined by suspension quantitative bactericidal test. Bacteria content in chicken house was determined by natural sinking method, and the effect of five disinfectants were also evaluated objectively through the on-the-spot disinfection of chicken house and disinfecting chicken house with chicken together.The result showed the MBC value of disinfectant E was smallest, but its bactericidal action was strongest, then disinfectant D, and bactericidal action of disinfectan C,B,A were weaker. After mixing 5 disinfectants with E.coli mixture for 1 min,5 min and 10 min, it was found that antimicrobial activity of disinfectant E to E.coli was higher than disinfectant C,B and A significantly (P﹤0.01), but there was significant difference between E and disinfectant D(P﹥0.05). And field disinfection trial in chicken house indicated that the effect of disinfectant E was superior to disinfectant D, and disinfectant E had significant effect to air, wall, ground, crib and waterer as well as cages surface, and disinfectant E influenced obviously mortality of chiken (P﹤0.05).After disinfecting, mortality of trialed chicken decreased 2.86%. So disinfectant E is safe and efficient, and it is fit to disinfect chicken house with chicken together,and it has good potential of application and popularization for disinfecting in livestock farm.
