调肝方药对愤怒和郁怒情绪模型大鼠不同脑区5-HTR2C表达水平的影响

【摘要】 目的:观察愤怒、郁怒情绪模型大鼠不同脑区中5羟色胺2C受体(5-HTR2C) mRNA和蛋白表达变化及调肝方药(经前平颗粒和经前舒颗粒)对其表达水平的调节作用,探讨愤怒、郁怒的发病机理,并探究调肝方药对大鼠愤怒、郁怒情绪的干预作用。方法:采用居住入侵加社会隔离的方法复制愤怒、郁怒情绪反应大鼠模型,以旷场实验和攻击行为测试评价模型,并应用调肝方药经前平、经前舒颗粒进行药物干预。造模后,将各组大鼠断头处死,分离顶区皮质、额区皮质、下丘脑和海马四个脑区。采用PCR基因扩增技术和蛋白印迹技术(Western blot)分别检测大鼠顶区皮质、额区皮质、下丘脑和海马脑区5-HTR2C mRNA和蛋白表达水平。结果:与正常组大鼠相比,愤怒情绪模型组大鼠5-HTR2C mRNA和蛋白表达水平在额区皮质、海马脑区均极显著性增高(P<0.01),在下丘脑脑区极显著性降低(P<0.01)。与愤怒模型组相比,经前平给药组大鼠5-HTR2C mRNA和蛋白表达水平在顶区皮质、额区皮质、海马脑区均极显著性降低(P<0.01),在下丘脑脑区极显著性增高(P<0.01)。与正常组大鼠相比,郁怒情绪模型组大鼠5-HTR2C mRNA和蛋白表达水平在下丘脑脑区极显著性降低(P<0.01),在海马脑区极显著性升高(P<0.01),在额区皮质脑区无显著性改变。与郁怒模型组相比,经前舒给药组大鼠5-HTR2C mRNA和蛋白表达水平在海马脑区极显著性降低(P<0.01),在额区皮质脑区无显著性改变。结论:大鼠不同脑区的5-HTR2C可能与大鼠愤怒、郁怒情绪密切相关,5-HTR2C表达变化可能是诱发大鼠愤怒、郁怒情绪的重要原因之一。调肝方药对愤怒、郁怒情绪大鼠不同脑区5-HTR2C有明显的调节作用,提示调肝方药对大鼠不同脑区5-HTR2C的调节作用可能是其干预大鼠愤怒、郁怒情绪的作用机制之一。

【Abstract】 Objective:Our aims are to study the mechanisms of anger emotion and the Liver-regulating Compound Recipe ( Jingqianping granule and Jingqianshu granule)acting on the anger-in and anger-out emotions,by investigating the expressions of 5-hydroxytryptamine 2C receptor(5-HTR2C) in different brain regions of anger-out or anger-in model rats,and the effects of Liver-regulating Compound Recipe ( Jingqianping granule and Jingqianshu granule)on the expression.Methods:Model rats with anger-out or anger-in emotional reaction are induced by social isolation plus resident intruder stress, evaluated by open-field test and aggressive behavior test,and some of them were intervened with Jingqianping(Jingqianshu)granule. After introducing model,we killed the rats by decapitation,dissected different brain regions(prefrontal cortex, parietal cortex, hippocampus and hypothalamus)。Then we analyzed the expressions of 5-HTR2C in partietal cortex,frontal cortex,hypothalamus and hippocampus of the three groups (including normal control group, anger-out or anger-in model group, Jingqianshu group or Jingqianping group) by the methods of RT-PCR and Western blot.Results: Compared with the normal control group,the mRNA and protein expressions of 5-HTR2C incressed obviously (P<0.01) in frontal cortex and hippocampus,decreased obviously (P<0.01) in hypothalamus of anger-out model rats.The mRNA and protein expressions of 5-HTR2C decreased obviously (P<0.01) in partietal cortex,frontal cortex and hippocampus, incressed obviously (P<0.01) in hypothalamus of the Jingqianping group compared with the anger-out model group.Compared with the normal control group,the mRNA and protein expressions of 5-HTR2C incressed obviously (P<0.01) in hippocampus,decreased obviously (P<0.01) in hypothalamus,and had no obviously changed in frontal cortex of anger-in model rats.The mRNA and protein expressions of 5-HTR2C decreased obviously (P<0.01) in hippocampus,and had no obviously change in frontal cortex of the Jingqianshu group compared with the anger-in model group.Conclusion:The expressions of 5-HTR2C in different brain regions were closely related to anger-out and anger-in of rats.The changes of the expressions may be one of the main reasons to induce rats anger-out or anger-in.The expressions of 5-HTR2C changed obviously in anger-out and anger-in model rats by using Liver-regulating Compound Recipe,so the regulating of Liver-regulating Compound Recipe to rats in different regions may be one of the mechanisms effecting the emotion of the anger-in or anger-out rats.
