六神丸通过Wnt信号转导通路抗肿瘤的临床与实验研究

【摘要】 目的:观察六神丸对非小细胞肺癌及人高转移肺腺癌细胞株A549的抑制作用,进一步探讨六神丸抗肿瘤的作用机理。方法:临床研究部分选取40例非小细胞肺癌患者随机分为观察组(GP方案+六神丸)和对照组(GP方案),其中观察组21例,对照组19例,两组均治疗2个疗程,比较两组治疗前后病灶大小、临床症状、体重、KPS评分以及毒副作用的变化情况。体外实验部分从研究Wnt信号通路入手,运用MTT、免疫组化和RT-PCR技术探讨六神丸可能存在的抑制肺癌细胞增殖的分子学机制。结果:临床研究证实,六神丸联合化疗可较好改善肿瘤患者大部分临床症状和一般情况,保护骨髓造血,减轻化疗所致的血液毒性,但在减小和稳定病灶、肝肾功的损害及消化系统毒副反应方面与单纯化疗相比无显著性差异。实验研究证实:细胞经六神丸含药血清干预24h后,药血清组OD值较正常鼠血清组增幅减慢,β-catenin蛋白的表达阳性率降低,促癌基因c-myc的表达含量下降。结论:六神丸通过阻断Wnt信号转导通路,实现其抗肿瘤作用,改善恶性肿瘤患者的临床症状,与化疗药物联用,可产生协同作用。

【Abstract】 Objective: To observe the supression of liu-shen Pill on non-small-cell lung cancer and lung cancer cells A549, and further to discuss the mechanism of liu-shen Pill on anti-tumor. Methods: In the clinical researchs, 40 patients with non-small-cell lung cancer patients were randomly divided into treatment group (GP regimen +liu-shen Pill) including 20 patients and the control group (GP regimen) including 19 patients. Changes of the tumor size, clinical symptoms, body weight, KPS score, and side effects between the two groups after two periods of treatment were observed. In the experimental studies, to explore the possible molecular mechanism of inhibiting the proliferation of lung cancer cells by neucleoproteinβ-catenin, the key factor of Wnt signaling pathway applying MTT, immunohistochemistry, and RT-PCR. Results: The clinical study confirmed that cancer patients of combination chemotherapy and liu-shen Pill can be much better in the clinical symptoms and general conditions, protect bone marrow to reduce the blood toxicity induced by chemotherapy, but in the stability and reduce lesions ,the impairment of liver and kidney function and toxicity of the digestive system compared with chemotherapy alone was no significant difference. In terms of experimental study confirmed: cells were treated with the serum containing liu-shen, the serum group has slower growth compared with normal mouse serum OD value, The positive rate ofβ-catenin,s expression of the serum group was lower than normal mouse serum, the serum group,s expression of cancer-promoting gene c-myc decreased.Conclusion: liu-shen Pill can improve the clinical symptoms of cancer patients, block Wnt signaling pathway to achieve its anti-tumor effect and combined with chemotherapy drugs, can produce synergy.
