奶牛中药复方“促孕散”化学成分提取工艺及活性研究

【摘要】 目的:通过渗漉法,利用非极性溶剂、极性溶剂、强极性溶剂依次提取中药“促孕散”的主要化学成分,通过测定提取物对小鼠子宫增重、卵巢发育影响及血清雌激素含量变化等活性试验得到其有效活性溶剂段,进而优化提取工艺,确定最佳提取条件,为中药提取技术研究以及减少临床用药量和发展中药新剂型提供理论依据。论文主要内容如下:首先以小鼠为受试动物,比较中药复方“促孕散”石油醚提取物、乙醇提取物、水提取物三种不同溶剂提取物对21日龄雌性小鼠子宫增重、卵巢卵泡发育、血清雌激素含量的影响来筛选中药复方“促孕散”主要活性成分。复方中药三种不同溶剂提取物中乙醇提取物对小鼠子宫增重、卵巢卵泡发育的影响极显著高于空白组(P<0.01);石油醚提取物组、水提取物与空白组差异不明显。乙醇提取物能明显促进小鼠子宫增重、卵巢卵泡发育和促使小鼠血清雌激素含量增加,表明中药复方“促孕散”主要活性成分为醇溶性成分。资料表明此方醇溶性成分主要为黄酮和生物碱。以淫羊藿苷含量为指标,采用超声提取技术,在单因素试验的基础上,通过正交试验确定“促孕散”中黄酮的最佳提取工艺。优选中药复方“促孕散”中黄酮的提取工艺及含量测定方法,确定以60%乙醇为溶剂,料液比1:12,超声提取2次,每次25min为中药复方黄酮最佳提取工艺;按最优工艺提取的“促孕散”中黄酮含量为1.236%。以盐酸水苏碱含量为指标,采用超声提取技术,在单因素试验的基础上,通过正交试验确定“促孕散”中生物碱的最佳提取工艺。优选中药复方“促孕散”中生物碱的提取工艺及含量测定方法,确定以60%乙醇为溶剂,料液比1:10,超声提取3次,每次20min为中药复方生物碱最佳提取工艺;按最优提取工艺提取“促孕散”生物碱得生物碱含量为1.148%。分别以黄酮提取物、生物碱提取物做动物实验筛选中药复方“促孕散”主要活性成分。结果表明:黄酮提取物对21日龄雌性小鼠子宫增重、卵巢卵泡发育和血清雌激素含量增加等的影响效果略高于生物碱提取物。表明黄酮为中药复方中主要活性成分。

【Abstract】 Major chemical composition of fertility-promoting intrauterine infusion powder(FPP) was extracted by percolation method and nonpolar solvent, polar solvent, powerful polar solvent was used in turns. The utility active solvent section was got by testing the increasing weight of the hystera seprately, the follicular diameter increasing and the content of the aquadiol in blood serum.Then the extraction technology was optimized and the best extraction condition was definited.All that had been done was to provid theory basing for the investigation of traditional Chinese drug extractive technique, reduce clinical drug consumption and develope new dosage. The article contents as follow:Major active component of chinese tradational medicine compound recipe FPP was screened by the effects of increasing the weight of the hystera and follicular development of the twenty-one days old feminity Mus musculus albus.Of course, the mouse was chosen as the first animal subject and compared the effects of three different solvent extract which contain ligroin extract, alcoho extract and water extract.Among three different solvent extractions, alcoho extract had exceeding significantly effects than the normal group on increasing the weight of the hystera and follicular development(P<0.01).There is no clear differenc between the ligroin extract group, water extract group and normal group.The effects of the alcoho extract showed that lyso-alcohol composition is the arch-active component.The data showed that the flavone and alkaloid were major in alcohol. Icariin was taken as the index, ultrasound-assisted extraction was used on the base of single factor experiment and the best extraction techniques of Flavanone component was selected by orthogonal experiment. The best extraction tec-niques:solvent:60% ethanol, extraction time:25 minitues, extraction times:2 times, and the solid-liquid ratio:1:12;the content of total flavonoids in FPP is 1.236%.Acid-stachydrine was taken as the index, ultrasound-assisted extraction was used on the base of single factor experiment and the best extraction techniques of alkaloidal component was selected by orthogonal experiment.The best extraction techniques:60% ethanol as solvent, solid-liquid ratio:1:10, extraction time:20 minitues, extraction times:3 times; the content of total alkaloids in FPP is 1.148%.The major activity component of FPP was chosen by the animal experiment which was fed the flavone extract and alkaloid extract.The results showed that the flavone extract had much better effects than the alkaloid extract on increasing the weight of the hystera and follicular development of the twenty-one days old feminity Mus musculus albus.All these showed the flavone are the major active component of the traditional Chinese medicine prescription.
