诺氟沙星配合物的制备及性能研究

【摘要】 诺氟沙星为第三代喹诺酮类药物,具有广谱、高效、低毒等特点。诺氟沙星与金属离子配位时,其结构中的羧基和羰基各提供一个氧原子与金属离子形成双齿配位配合物。本文选择两种具有重要生物功能的金属离子-Ni(Ⅱ)、Co(Ⅱ),选择合适的条件合成出两种新的金属诺氟沙星配合物,研究了它们的结构及这些金属诺氟沙星配合物的生物活性。1.以诺氟沙星为配体,合成了以Ni(Ⅱ)为中心离子的配合物,通过红外,紫外,电导率,X-射线单晶衍射,电化学等方法对配合物的组成和性质进行分析,结果表明为一新的配合物。2.以诺氟沙星为配体,合成了以Co(Ⅱ)为中心离子的配合物,通过红外,紫外,电导率,X-射线单晶衍射,电化学等方法对配合物的组成和性质进行分析,结果表明为一新的配合物。3.采用紫外可见光谱法研究了镍、钻-诺氟沙星配合物与DNA的作用。可以发现随着DNA浓度的增大,紫外光谱中出现减色效应,但红移现象表现不明显,通过计算这两种金属诺氟沙星配合物与DNA之间的结合常数来说明它们之间存在着部分插入作用。4.结合镍-诺氟沙星配合物特殊的“U”形结构,采用紫外可见光谱法研究了它对十四种苯类物质的选择吸附性。通过计算它们的最大减色率,说明该配合物对这十四种苯类物质存在着选择吸附。

【Abstract】 Norfloxacin is the third drugs of quinolones which have features of excellent activity, wide spectrum and low tociticy. Norfloxacin can bind with metal ions by the oxygen atom of the carboxyl group and carbonyl group. In this paper, some biological metal ions, such as Ni(Ⅱ), Co(Ⅱ), were selected to synthesize norfloxacin-metal complexes.1. A new Norfloxacin- Ni(Ⅱ) complexes were synthesized. Many techniques were used to analysis the structural properties such as infrared absorption spectroscopy, UV spectrometry, conductivity measurements, X-ray monocrystal analysis etc.2. A new Norfloxacin- Co(Ⅱ) complexes were synthesized. Many techniques were used to analysis the structural properties such as infrared absorption spectroscopy, UV spectrometry, conductivity measurements, X-ray monocrystal analysis etc.3. UV spectrometer was used to study the interactions of the two metal complexes with DNA. It was found that, as the DNA concentration increases, hypo chromic effect in UV spectrometer can be observed, but the red-shift in not obvious, which indicates that norfloxacin-metal complexes partly inserted into DNA. The binding constants of metal complexes with DNA were calculated and the interaction mode was discussed.4. UV spectrometer was used to study the interactions of the Nickel(II) complexes with fourteen substances of benzene kinds. It is found that, The complexes alternative enroll these substances of benzene kinds by counting their maximum hypo chromic effect.
