复方血塞通滴丸指纹图谱与药代动力学研究

【摘要】 复方血塞通滴丸是由三七、葛根与山楂提取、纯化的有效部位,滴制而成的现代中药制剂。具有治疗中风偏瘫、淤血阻络证的作用,主治动脉粥样硬化性血栓性脑梗塞、脑栓塞等。本实验首先采用高效液相色谱法,以葛根素、芦丁为参照峰,分析10批样品,确定了22个共有峰,并采用《中药色谱指纹图谱相似度评价系统(国家药典委员会2004A版)》评价了指纹图谱的相似度,建立了该制剂的HPLC指纹图谱;然后对通过单味药材,阴性对照液及对照品与全方指纹图谱的比较研究,对全方22个共有峰进行了归属分析,并对其中5个色谱峰进行了确认,明确了该方的药效物质基础。本文还对复方血塞通滴丸进行了药代动力学研究,采用高效液相色谱法,以三七中人参皂苷Rg1、人参皂苷Rb1为检测指标,分别测定灌胃给予复方血塞通滴丸、三七总皂苷粉、三七与葛根配伍组、三七与山楂配伍组后大鼠血浆中人参皂苷Rg1、人参皂苷Rb1的血药浓度变化规律,考察两种指标成分在大鼠体内的吸收。所得数据经DAS2.1.1软件拟合均为二室模型。实验结果表明,全方配伍后人参皂苷Rg1的AUC、Cmax与t1 / 2β值均高于单味三七总皂苷;人参皂苷Rb1的Cmax、AUC值也均高于单味三七总皂苷,证实了复方滴丸的作用优于单味药,具有提高疗效的作用,为复方血塞通滴丸的临床应用以及从药代动力学角度阐明方剂配伍的合理性与整体性提供了科学依据。

【Abstract】 Co-Xuesaitong Dropping Pill is an effective preparation to cure stagnation of blood . It consists of ingredients of Fructus crataegi, Radix puerariae and Panax pseudoginseng.In this paper, we firstly established the HPLC fingerprint of Co-Xuesaitong Dropping Pill and selected 22 fingerprint peaks.All the peaks were well reproducible, special to Co-Xuesaitong Dropping Pill and they could reflect Co-Xuesaitong Dropping Pill quality. Performed by HPLC to identify the chemical constituents of the whole and individual herb of Co-Xuesaitong Dropping Pill, 5 peaks were identified.We also studied the pharmacokinetics of Rg1 and Rb1 from Co-Xuesaitong Dropping Pill in rats, and compare the pharmacokinetics of Rg1 and Rb1 after ig administration of Co-Xuesaitong Dropping Pill ,panax notoginseng saponins,panax notoginseng compatibility with kudzuvineroot and panax notoginseng compatibility with hawthom fruit .Rg1 and Rb1 that is double hump absorption is two -compar- tment model.There existed significant differences in the values of elimination AUC, Cmax and t1/2βof Rg1 , Cmax, AUC of Rb1 among Co-Xuesaitong Dropping Pill and panax notoginseng. The results showed that Rg1 and Rb1 were absorbed more quickly after administration of Co-Xuesaitong Dropping Pill than panax notoginseng. The compound compatibility of the decoction promoted the absorption of Rg1 and Rb1. It indicated that there is rationality in the compatibility of Co-Xuesaitong Dropping Pill .
