基于因子分析的附子理中方对脾虚泄泻的治疗作用及关键因子研究
Effect and key factors study of FuziLizhong prescription on Pixuxiexie syndrome based on factor analysis
【摘要】 目的:研究附子理中方对脾虚泄泻大鼠模型的治疗作用,同时采用因子分析法分析其治疗环节的关键因子。方法:大黄水煎液灌胃30 d建立脾虚泄泻大鼠模型,以附子理中方干预21天后,酶联免疫吸附(ELISA)法检测各组大鼠血清、血浆和组织中血管活性肠肽(VIP)、胃泌素(GAS)、胃动素(MTL)、白细胞介素1(IL-1)、白细胞介素6(IL-6)、肿瘤坏死因子α(TNF-α)、干扰素γ(IFN-γ)、超氧化物歧化酶(SOD)和丙二醛(MDA)等指标的含量,同时通过因子分析方法综合分析提取附子理中方治疗脾虚泄泻的关键因子。结果:附子理中方能明显降低大鼠稀便率,在胃肠激素调节方面,具有升高GAS、MTL,降低VIP的作用,在免疫细胞因子的调节方面,具有降低IL-1、IL-6、TNF-α、IFN-γ的作用,在自由基的调节方面,具有降低MDA含量,维持SOD活性的作用。通过因子分析,从9个指标中抽提出2个公因子,共解释信息占92.61%,同时构建了脾虚泄泻模型综合评价函数。结论:附子理中方具有减少胃肠液分泌、保护胃黏膜、减轻炎症反应、减少自由基生成的作用。免疫因子和胃肠激素因子为附子理中方调节脾虚证的关键因子。对附子理中方不同剂型的作用进行比较发现,长期给药后,丸剂与汤剂在以上作用无明显差异,但丸剂在IFN-γ的调节上,效果优于汤剂。因子分析可以对脾虚泄泻模型进行综合评价,是一种适宜中医动物模型的评价方法。
【Abstract】 Objective: To study the therapeutic effect and key factors of FuziLizhong prescription on Pixuxiexie rats by factor analysis method. Methods: Pixuxiexie rat model was established by oral administration of Rhubarb decoction for 30 days. After the model was established, FuziLizhong prescription was administered for 21 days. The contents of VIP, GAS, MTL, IL-1, IL-6, TNF-α, IFN-γ, SOD and MDA in serum, plasma and tissues of rats in each group were detected by ELISA. At the same time, the key factors of treating Pixuxiexie syndrome were analyzed by factor analysis. Result: FuziLizhong prescription decreased the diarrhea rate in rats, it also increased gastrointestinal hormone related factors GAS, MTL and decreased VIP. It reduced immune cell cytokines IL-1, IL-6, TNF-α and IFN-γ. It also reduced the content of free radicals MDA and maintained SOD activity. Two common factors were extracted from nine indexes by factor analysis, which explained 92.613% of the information and a comprehensive evaluation function of Pixuxiexie model was constructed. Conclusion: FuziLizhong prescription has the effects of reducing gastrointestinal secretion, protecting gastric mucosa, alleviating inflammation and reducing free radical production. Immunological factor and gastrointestinal hormone factor are the key factors of regulating Pixuxiexie syndrome. Comparing the effects of different formulations of FuziLizhong prescription,it shows no significant difference between pills and decoctions after long-term administration, but the effect of regulating IFN-γ is better in dosage form of pills than decoctions. Pixuxiexie model can be comprehensively evaluated by factor analysis, which may be a suitable evaluation method for animal models of traditional Chinese medicine.
