根系活力检测中保险粉用量的确定

【摘要】 根系活力是常用的反映植株生活力的指标,TTC(2,3,5-三苯基氯化四氮唑)还原法是使用最广泛的方法。但现有研究论文及植物生理学实验指导所介绍的方法中保险粉用量缺乏合理性。本研究旨在分析这种不合理性是否误导现有根系活力相关的研究,并通过试验确定适宜保险粉用量。采用文献计量法对现有文献中的根系活力数值进行分析。保险粉用量试验从0.02~0.20g共7个不同量,各加5mL 5mg·mL-1TTC溶液,生成三苯基月替(TPF)后加乙酸乙酯10mL,研究保险粉用量与日立紫外分光光度计(U-3900H)485nm下吸光值的关系,以确定合适用量。文献计量分析结果表明,现有根系活力测定结果的偏差超出合理范围。试验结果显示,保险粉的用量小于150mg时,与吸光值呈线性相关,超过150mg后,吸光值不再增加。因此,5mL TTC溶液中加入保险粉的最低量应为150mg。

【Abstract】 The use of 2,3,5-triphenyltetrazolium chloride(TTC)test for determining roots viability is apopular method.However,existing root viability methods described in research articles or experiment manuals in plant physiology cannot give a suitable amount of sodium hydrosulfite.In this case,an experiment was designed to to investigate the appropriate usage of sodium hydrosulfite.Seven different amounts of sodium hydrosulfite ranging from 0.02 to 0.20 g were arranged to reduce 5 mL5 mg·mL-1 TTC.Carmine red TPF(1,3,5-triphenylformazan)was extracted by 10 mL ethyl acetate.The extract was tested using the U-3900 HSpectrophotometer at 485 nm.The data of root viability extracted from existing publications were also analyzed using bibliometric method.The results of bibliometric analysis showed that bias of existing data doesn’t make sense the accuracy of currently available data in the literature.A significant positive correlation was found between the amount of sodium hydrosulfite and absorbance belowe 150 mg,whereas the absorbance remained constant for over150 mg of sodium hydrosulfite.A minimum of 150 mg sodium hydrosulfite is required to construct a reliable standard curve.Finally,a reasonable method of preparing standard curves was recommended.
