解磷菌的培养条件优化研究
Culture Conditions Optimization Study of Phosphate-solubilizing Bacteria
【摘要】 近年来的研究表明,解磷微生物能够将土壤中的难溶性磷转化为植物可以吸收并利用的可溶性磷,解磷菌制剂可以有效提升土壤肥力,减少化肥使用量。本文通过选择适合解磷菌株的培养条件,增加菌株的解磷能力和繁殖能力。根据钼锑抗比色法测定不同培养条件下菌株的解磷能力,结合测得的菌体生长量确定最佳培养条件。结果表明,该解磷菌株在28℃,pH 6.5,以葡萄糖作为碳源,以硫酸铵作为氮源,摇床速率180r/min,Mg2+、Mn2+离子的浓度为0.03g/L,可达到最佳培养条件。培养条件经过优化后,磷浓度可达0.780mg/L。
【Abstract】 Recent studies have shown that phosphate-solubilizing microorganisms can effectively transform insoluble phosphorus in soil into soluble phosphorus which can be absorbed and utilized by plants. The preparation of phosphate-solubilizing bacteria can effectively improve soil fertility and reduce the use of chemical fertilizer.In this paper, the suitable conditions of culturewere selected to increase the ability of phosphorus release and reproduction of the strain.According to the molybdenum antimony colorimetric method, the phosphorus releasing ability of the strain was determined under different culture conditions, and the optimum conditions of culture were determined by combining with the measured growth of bacteria.The results showed that at 28 ℃, pH 6.5, glucose as carbon source, ammonium sulfate as nitrogen source, shakerspeed of 180 r/min and 0.03 g/L concentration of magnesium ion manganese ion, the optimum conditions of culture could be achieved. After the culture conditions were optimized,phosphorus concentration can reach 0.780 mg/L.
