大孔树脂纯化芹菜籽黄酮的工艺

【摘要】 芹菜籽的药理作用非常广泛,有较高的使用价值.黄酮类化合物是一类广泛存在的天然抗氧化剂,具有多种生理功能.本文利用大孔树脂纯化芹菜籽黄酮,以回收率为指标进行正交实验,结果表明:大孔树脂纯化芹菜籽黄酮的最佳纯化工艺为:上样液浓度8. 01 mg·L-1、上样液pH值4. 8、上样液速率2. 5 m L·min-1、洗脱液速率1 m L·min-1、洗脱液体积分数87%.在该条件下,纯化样品中的黄酮纯度由2. 12%提高到21. 7%,提高了9. 2倍,总黄酮回收率达到94. 9%.因此,大孔树脂纯化芹菜籽黄酮类化合物效果较好.

【Abstract】 Celery seed is widely used as food and medicine as well. Flavonoids are a kind of natural antioxidants with a wide range of physiological functions. In the curren tresearch,as celery seed flavonoid was purified by macroporous resin,orthogonal experiments were carried out with the recovery as the index to determine the optimal purification process,during which the concentration of sample solution was 8. 01 mg·L-1,the pH value of sample solution 4. 8,the rate of sample solution 2. 5 m L·min-1,the rate of eluent 1 mL·min-1,and the elution liquid integral number 87%. Under the aforementioned condition,the purity of flavonoids in the sample increased from2. 12% to 21. 7% —9. 2 times higher,and the total flavonoid recovery reached 94. 9%. It is concluded that macroporous resin is suitable for separating and purifying celery seed flavonoid.
