马铃薯疮痂病拮抗菌Bacillus amyloliquefaciens12-82发酵条件优化
OPTIMIZATION OF FERMENTATION CONDITIONS OF Bacillus amyloliquefaciens 12-82 ANGAINST TO POTATO SCAB
【摘要】 马铃薯疮痂病(Potato scab)是由链霉菌(Steptomyces spp.)引起的重要病害。前期工作中筛选到一株对马铃薯疮痂病菌具有较强拮抗活性的菌株Bacillus amyloliquefaciens 12-82。本研究通过响应面优化方法对该拮抗菌产芽孢条件进行优化,以满足生产的需要。结果表明,当培养基组成为2%玉米粉、2%蛋白胨、0.5%CaCl2;培养条件为发酵时间48 h,装瓶量50 mL/150 mL,接种量10%,初始pH值7.3,培养温度33℃,芽孢数量可达到3.1×108 CFU/mL,实验结果对该菌株的开发应用具有一定指导意义。
【Abstract】 Potato scab is an economic disease caused by Steptomyces spp. In previous work, a strain named Bacillus amyloliquefaciens 12-82, which had a good antagonistic resistance to potato scab, was screened. In order to meet the needs of production, this study optimized the spore production conditions of the antagonistic bacteria using response surface optimization. Results showed that the medium consisted of 2% cornmeal, 2% peptone and 0.5% CaCl2, culture conditions were fermentation time 48 h, bottle filling amount to 50 mL/150 mL, inoculation quantity 10%, initial pH 7.3, culture temperature 33℃, the spore number was 3.1×108 CFU/mL, the experimental results will provide certain guiding significance for the development and application of the strain.
