江苏省6个翘嘴鳜群体的遗传多样性分析

【摘要】 利用20对微卫星引物对江苏省5个内陆湖(太湖、滆湖、洪泽湖、高邮湖和骆马湖)和长江的翘嘴鳜(Siniperca chuatsi)野生种群进行遗传多样性分析,研究江苏省不同水域翘嘴鳜的遗传多样性差异和亲缘关系。结果表明,6个水域的翘嘴鳜群体遗传多样性丰富,其中长江翘嘴鳜群体的平均有效等位基因数为6. 524 8,平均期望杂合度为0. 922,平均多态信息含量为0. 827 3,均高于其它5个翘嘴鳜群体,说明长江翘嘴鳜群体的遗传多样性高于其它5湖的翘嘴鳜群体。哈迪温伯格平衡显示,只有2个位点上存在不平衡,其余都满足哈迪温伯格平衡。从6个水域翘嘴鳜群体之间的遗传相似系数和遗传距离来看,太湖翘嘴鳜群体和滆湖翘嘴鳜群体的亲缘关系最近,骆马湖翘嘴鳜群体和太湖翘嘴鳜群体的亲缘关系最远。研究结果可为江苏省翘嘴鳜种质资源的保护、监测和遗传育种提供分子基础资料。

【Abstract】 20 pairs of microsatellite primers were used to analyze the genetic diversity of the wild population of Siniperca chuatsi in 5 inland lakes( Taihu Lake,Gehu Lake,Hongze Lake,Gaoyou Lake and Loma Lake) in Jiangsu Province and in the Yangtze River,the genetic diversity differences and phylogenetic relationships of Siniperca chuatsi in different waters were also analyzed. The results showed that the genetic diversity of Siniperca chuatsi populations in the waters was very rich. The average effective allele number of Siniperca chuatsi population in the Yangtze River was 6. 524 8,the average heterozygosity was 0. 922 and the average polymorphism information content was 0. 827 3. The three indexes were higher than the other five species,indicating that the genetic diversity of Siniperca chuatsi population of the Yangtze River was higher than that of other five Siniperca chuatsi populations. Hardy Weinberg equilibrium showed that only two loci were unbalanced,and the others complied with Hardy Weinberg equilibrium. From the genetic similarity coefficient and genetic distance among the 6 populations of Siniperca chuatsi,the relationship between Taihu Lake population and Gehu Lake population was the closest,and the relationship between Taihu Lake population and Luoma Lake polupation was the farthest. The results of this study provide molecular basis for monitoring,protection and genetic breeding of Siniperca chuatsi germplasm resources in Jiangsu Province.
