以Lorke法为基础的半致死剂量分析法在油酸和化香树果序及黄连素抗镉减毒评价中的应用
Application of LD50 Analysis Method in Evaluation of Oleic Acid,Platycarya strobilacea Caryopsis and Berberine on Resistance and Attenuation of Cadmium Toxicity Based on Lorke Method
【摘要】 镉污染严重威胁着农业的健康发展,如何有效抗镉减毒是保障农业生产安全的重要措施。然而,目前缺乏高效的抗镉药物及简便易行的评价方法。本试验借助Lorke法为基础的半致死剂量(LD50)分析方法 ,检测了油酸、化香树果序、黄连素等不同潜在的抗镉化合物的减毒作用。通过改良后的Lorke法测定不同抗镉药物对小鼠的经口镉LD50的影响。结果表明,与200 mg/kg VC相比,灌胃200 mg/kg油酸、化香树果序、黄连素能够减轻镉的毒性,提高镉的LD50。LD50由140~160 mg/kg分别上升至300~320 mg/kg(油酸组)、280~300 mg/kg(化香树果序组)和320~340 mg/kg(黄连素组)。以上结果表明,以Lorke法为基础的LD50分析方法,能够评价出油酸、化香树果序、黄连素的减镉毒作用及其剂量-效应关系。以上结果同时证明了相同剂量下黄连素效果要优于油酸、优于化香树果序。
【Abstract】 Cadmium pollution is a serious threat to the development of agriculture,control and reduction of cadmium is an important way to ensure the safe of agricultural production.However,effective methods for controlling and evaluating cadmium toxicity are not available currently.To effectively reduce the toxic and adverse effects caused by cadmium,the LD50 analysis method based on Lorke method was used to evaluate the attenuating effect of oleic acid,Platycarya strobilacea caryopsis and berberine on cadmium toxicity.The modified Lorke method was used to determine the effects of different anti-cadmium drugs on transoral cadmium LD50 in mice.The results showed that 200 mg/kg of oleic acid,P.strobilacea caryopsis and berberine could reduce the toxicity of cadmium and increase the LD50 of cadmium in mice compared with 200 mg/kg of vitamin C.Their LD50 of cadmium rose to300-320 mg/kg,280-300 mg/kg and 320-340 mg/kg,respectively.The results indicated that the LD50 analysis based on Lorke method was feasible to evaluate the attenuating effect of oleic acid,P.strobilacea caryopsis and berberine on cadmium toxicity,as well as the relationship between dose and effect.The results also proved that the attenuating effect of berberine on cadmium toxicity was better than that of oleic acid and P.strobilacea at the same dose.
