【摘要】 目的:研究活血通络汤在防治下肢深静脉血栓形成中的作用机理,主要从保护血管内皮细胞这一角度来探讨活血通络汤在下肢深静脉血栓形成中的干预机制,为临床治疗提供客观依据。方法:通过结扎下腔静脉下端与双侧髂总静脉建立大鼠深静脉血栓形成模型,研究活血通络汤对大鼠一般状态和双后肢肿胀情况,静脉血栓形成情况,炎症反应情况,血管内皮细胞损伤情况等的影响。通过建立二甲苯所致小鼠耳廓肿胀急性炎症模型,研究活血通络汤的对耳廓肿胀度的抑制作用。通过H₂O₂诱导人脐静脉内皮细胞建立体外血管内皮细胞氧化损伤模型,以及通过建立体外血管内皮细胞缺糖损伤模型,进行细胞形态学观察,LDH、NO和MDA的测定,细胞凋亡检测,研究活血通络汤对血管内皮细胞的保护作用。结果:在大鼠深静脉血栓形成模型实验中,模型组大鼠的一般状态,双后肢肿胀情况以及静脉血管病理形态学改变较假手术组程度重,其炎细胞浸润,血管内皮损伤严重。活血通络汤组大鼠的一般状态,双后肢肿胀情况以及静脉血管病理改变均有所改善。在小鼠耳廓肿胀急性炎症模型实验中,活血通络汤具有抑制耳廓肿胀度的作用。在体外血管内皮细胞氧化损伤、缺糖损伤模型实验中,活血通络汤用药组多数细胞形态没有出现明显的改变,细胞数量没有明显减少现象,LDH、MDA含量与模型组比较有明显减小,活血通络汤干预后的HUVEC凋亡小体数量有较明显的减少。在体外血管内皮细胞缺糖损伤模型实验中,活血通络汤用药组整体的细胞生长情况好于模型组。结论:静脉结扎法可以有效建立深静脉血栓形成模型。深静脉血栓形成与血管内皮细胞损伤有相关性。活血通络汤可以抑制炎细胞浸润,保护血管内皮细胞,减少血栓形成,能清除氧自由基,减少细胞凋亡,对氧化损伤诱导的血管内皮细胞具有保护作用,对缺糖损伤诱导的血管内皮细胞具有保护作用。活血通络汤的作用效果与剂量有一定的量效关系。活血通络汤剂量越高,作用效果越明显。中药可能通过保护血管内皮细胞达到防治下肢深静脉血栓形成的作用。更多还原

【Abstract】 Objective: To research the mechanism of Huo Xue Tong Luo Broth in preventing deep venous thrombosis, mainly from the view of the protection of endothelial cells to study on interventional mechanism of Huo Xue Tong Luo Broth in deep venous thrombosis, in order to provide an objective basis for clinical treatment.Methods: Deep venous thrombosis model in rats was established through ligation of inferior vena cava and bilateral ligation of common iliac vein.The role of Huo Xue Tong Luo Broth was studied through the mouse ear model of acute inflammation and vascular endothelial cell injury model.Results: Huo Xue Tong Luo Broth can inhibit thrombus formation in rats (p<0.01), degree of inhibition of mouse ear edema (p<0.05), protect HUVEC cells which induced by glucose deprivation injury and oxidation injury.Conclusions: Vein ligation can effectively set up the model of deep vein thrombosis. Deep vein thrombosis and vascular endothelial cell injury has relevance. Huo Xue Tong Luo Broth can inhibit inflammatory cell infiltration, protect vascular endothelial cells, reduce thrombosis, removal of oxygen free radicals and reduce cell apoptosis. Huo Xue Tong Luo Broth can protect vascular endothelial cells which induced by glucose deprivation injury and oxidation injury. By protecting vascular endothelial cells , Chinese herbs could achieve the prevention and treatment of lower extremity deep venous thrombosis of the purpose.更多还原
