加热致小鼠睾丸生精障碍相关蛋白质及其功能的研究

【摘要】 精子发生是一个连续的细胞分裂分化过程,包括精原细胞分裂增殖、精母细胞减数分裂和精子形成三个主要阶段。精子发生过程涉及染色体数目的减半、遗传重组、染色质重塑及浓缩、精子顶体和尾部的形成等复杂的变化过程,所有这些高度有序的过程均需要相关蛋白质进行精确地调节。分离和鉴定与精子发生相关的蛋白质并探讨其相关功能对于揭示男性生殖的分子调控机制及临床男性不育症的诊治具有重要的理论和实践意义。 为了研究小鼠睾丸局部短暂加热导致小鼠生精障碍的分子机制,我们运用蛋白质组学研究常用的技术手段—双向凝胶电泳(two-dimensional electrophoresis,2-DE)和质谱(mass spectrometry,MS)分离和鉴定小鼠睾丸加热后发生改变的蛋白质。首先我们创建了含有小鼠睾丸双向凝胶电泳参考胶的符合联盟标准的数据库,在参考胶上有504个通过肽质量指纹谱鉴定的蛋白点,点击胶上的蛋白点可链接到对应的蛋白质条目,同时还可得到蛋白质的鉴定信息,并可链接到SWISS-PROT或NCBI蛋白质网络数据库,另外,数据库支持复杂的查询功能,该公共数据库为研究正常小鼠睾丸功能和小鼠睾丸病理提供了有价值的资源;其次,我们采用类似的方法分析了小鼠睾丸局部
【Abstract】 Spermatogenesis is the process by which stem cells develop into mature spermatozoa. There are three phases: (1) Spermatocytogenesis (Mitosis), (2) Meiosis, and (3) Spermiogenesis. Many testis-specific proteins are required for mitotic proliferation of spermatogonia, spermatocytes undergoing genetic recombination and meiotic divisions, and differentiation of haploid spermatid. Isolation and identification of spermatogenesis related proteins and further functional study are very important to the elucidation of molecular mechanism involving in meiosis and spermiogenesis. It is also important to clinical diagnosis and treatment of male infertility and male molecular contraception.We characterized cellular and molecular mechanisms involved in spermatogenesis following short-term heat exposure of murine testis. For these studies we utilized a proteomic approach with two-dimensional gel electrophoresis (2-DE) analyses and mass spectrometry (MS)
