电针对老年性痴呆大鼠脑内生化及学习记忆信号转导机制的影响
The Study of the Effect of Electro-Acupuncture on the Learning and Memory Signal Transduction Mechanism and Brain Biochemistry in Laboratory Rats with Senile Dementia

【摘要】 目的:探讨电针对老年性痴呆大鼠学习记忆影响的机理。 方法:将老年痴呆(AD)大鼠分为空白组、假损伤组、模型组和电针组,采用D-半乳糖结合氯化铝损害模型的造模方法,选“百会”、“大椎”、“太溪”、“足三里”电针治疗。分别用生化方法观察电针对AD大鼠脑组织胆碱酯酶、超氧化物歧化酶、单胺氧化酶、一氧化氮、一氧化氮合酶的影响;用免疫组化方法观察海马组织蛋白激酶的表达;用光、电镜方法观察大鼠脑组织形态结构的变化;用原位杂交方法观察海马组织天门冬氨酸受体rnRNA的表达;用水迷宫和电迷宫实验观察大鼠逃避潜伏期和错误次数,以了解其学习记忆能力。 结果:电针可明显降低痴呆模型组大鼠脑组织AchE活力;电针提高了SOD的活性,抑制MAO活性,提高了抗氧化系统的能力;电针能提高模型大鼠海马匀浆中NO、NOS含量,提高了大鼠学习记忆能力;电针通过调节蛋白激酶、天门冬氨酸受体rnRNA的表达影响长时程增强(LTP)的形成;电针能影响海马突触形态;电针能使AD大鼠在水迷宫和电迷宫实验中逃避潜伏期和错误次数减少,使AD大鼠学习记忆能力得到一定程度的恢复。电针不失为治疗老年性痴呆的简单、有效的方法。
【Abstract】 Objective: To explore the mechanism of electro-acupuncture effect on senile dementia rat’s learning and memory function.Methods: Divide senile dementia AD rats into normal control group, sham-operation group, model group and electro-acupuncture group randomly, adopt D-galactoside combinated aluminum chloride damage model, treat Baihui, Dazhui, Taixi and Zusanli with electro-acupuncture. Use biochemical method to observe the effect of electro-acupuncture on AchE, SOD, MACK NO, NOS of senile dementia AD rat’s brain tissue; use immunohistochemistry technique to observe protein kinase expression of hippocampal; use optical and electron microscope to observe morphosis change of rat’s brain tissue; use in situ hybridism technique to observe aspartate receptor mRNA expression of hippocampal; use water and electric maze experiment to observe rat escape latent period and mistake times, to understand it’s learning and memory ability.Results: Electro-acupuncture can decrease AchE ability of VD rat’s brain tissue evidently; electro-acupuncture can increase ability of SOD , inhibit ability of MAO , increase ability of antloxidation system; electro-acupuncture can increase model rat’s NO, NOS content of hippocampal homogenate, increase learning and memory ability of rat ; electro-acupuncture can influence formation of long-term potentiation ( LTP) by regulate protein kintasc, aspartate receptor mRNA expression. Electro-acupuncture can influence morphosis of hippocampal synapse. Electro-acupuncture can decrease AD rat escape latent period and mistake times in water and electric maze experiment, improve AD rat’s learning and memory in some extent. Electro-acupuncture is a simple and effective method for treating VD.
