鹅绒藤生药学及化学成分研究

【摘要】 目的:对鹅绒藤进行全面系统地生药学研究,并进行化学成分提取分离,初步建立起鹅绒藤药材质量标准。方法:采用生药学研究方法研究鹅绒藤原植物、药材性状、不同药用部位的组织构造及其粉末特征;采用扫描电镜技术对鹅绒藤地上部分的各个部位进行微形态观察;采用红外三级鉴定方法研究鹅绒藤药材、不同药用部位、药材不同极性溶剂提取物以及不同产地药材的红外光谱;采用《中国药典》方法进行纯度检查和浸出物含量测定;采用高相液相色谱法研究鹅绒藤指纹图谱并测定山柰苷含量;采用系统分离法研究鹅绒藤药材乙醇提取物的化学成分。结果:确定了鹅绒藤基原、性状、显微鉴别和微形态的专属性特征;制定了鹅绒藤红外对照指纹图谱;建立了鹅绒藤的HPLC对照指纹图谱;鉴定出6个化合物,分别是十四烷酸甘油酯、二十八烷醇、20R-n-Butylpregn-5-en-3β-ol、胡萝卜苷、羽扇豆醇和山柰苷。其中化合物20R-n-Butylpregn-5-en-3 p-ol、胡萝卜苷、羽扇豆醇、十六烷酸甘油酯、二十八烷醇是首次从鹅绒藤中分离得到;初步拟定了鹅绒藤药材的质量标准草案。结论:为鹅绒藤的生药鉴定、质量控制、质量标准的制定、化学成分分离鉴定、药理活性研究以及合理开发应用提供了系统的科学资料,为鹅绒藤的深入研究奠定了坚实基础。

【Abstract】 Object:In order to establish the quality standard of Cynanchum chinense R.Br., a systematic pharmacognostic research and isolation of chemical constituents of Cynanchum chinense R.Br. were performed.Methods:Pharmacognostic researches aimed at the botanical resource, medical character and microstructure of Cynanchum chinense R.Br. Scanning electron-microscope was applied in the micro-morphological research on its aerial part. Three-Steps Infrared Macro-Fingerprint Method was used to study the infrared fingerprint of Cynanchum chinense R.Br. from different area, of different parts, and of abstraction with different polarity solvent. Spectrophotometry was used to determine the content of total flavone. Its fingerprint was compared by applying High-Performance Liquid Chromatography (HPLC) to determine the concentration of kaempferitrin. Its chemical components of alcohol extractive were analyzed using systematic separation process. The test on its purity and the content of extract was performed using the methods from Chinese Pharmacopoeia.Results:This research determined the traits in the origin, character, and micro-morphology of Cynanchum chinense R.Br., and also determined the fingerprint of the infrared and HPLC. Besides, this research established the content standard of water, ash, extract, flavone and kaempferitrin.6 compounds were identified from Cynanchum chinense R.Br. and 20R-n-Butylpregn-5-en-3β-ol was isolated from botanical resource for the first time.4 compounds, daucosterol, lupeol, hexadecanoic acid glyceride and octacosanol, were isolated from Cynanchum chinense R.Br. for the first time and identified. The content of different inorganic elements and amino acid were analyzed. The standard for crude drug quality of Cynanchum chinense R.Br. was proposed initially. Conclusions:This research summarized systemic scientific data, and established the foundation for the pharmacognosy, quality control, quality standard, isolation and identification of chemical components, pharmacological activity of Cynanchum chinense R.Br., which is definitely helpful for further research on Cynanchum chinense R.Br.
