乳岩宁方下调MCF-7荷瘤裸鼠HER-2基因表达及提高免疫功能的实验研究

【摘要】 目的:通过观察乳岩宁方对MCF-7乳腺癌荷瘤裸鼠HER-2/nue基因表达及对免疫功能的影响,探讨其对乳腺癌的抑瘤作用,并初步研究其作用机制。材料与方法:将人乳腺癌细胞株MCF-7接种于BALB/C-nu/nu裸鼠右侧胸壁第二乳垫部皮下,选取荷瘤成功裸鼠随机分为对照组、乳岩宁组、平消胶囊组,每组9只。记录各组裸鼠精神状态、饮食、体重变化情况;第18天,处死裸鼠取脾、肿瘤,称重,计算脾指数、抑瘤率;制作透射电镜切片,透射电镜下病理形态学观察;Western Blotting检测肿瘤组织中HER-2/nue蛋白的表达。结果:1.乳岩宁方能改善荷瘤裸鼠生存质量,维持荷瘤裸鼠的体重,乳岩宁组体重净降明显低于对照组、平消胶囊组(p﹤0.01)。2.乳岩宁组和平消胶囊组的瘤重均低于对照组,与对照组相比差异性显著(p﹤0.01)。3.乳岩宁组的脾指数最高,与平消胶囊组相比差异性显著(p﹤0.05),与对照组相比无差异性显著(p>0.05)。4.透射电镜观察结果显示乳岩宁方能够诱导MCF-7乳腺癌细胞凋亡。5. HER-2/nue蛋白表达结果:乳岩宁组、平消胶囊组均低于对照组,与对照组比较差异性显著(p﹤0.05)。结论:1.乳岩宁方能改善MCF-7荷瘤裸鼠机体状况,提高生活质量。2.乳岩宁方能提高MCF-7荷瘤裸鼠脾指数,可能提高了裸鼠的免疫功能。3.乳岩宁方具有一定的抑瘤作用,可能通过诱导肿瘤细胞凋亡和抑制肿瘤组织中HER-2/nue基因蛋白的表达来实现的。
【Abstract】 Purpose: The aim of this study is to observe the influence of Ruyanning Decoction to HER-2 gene and immune function in MCF-7 mammary cancer loaded on Nude mice and to investigate the tumor-inhibition function and action mechanism.Material and method:BALB/C Nude mice are injected MCF-7 on the second breast pad of the right chest wall subcutaneously .Twenty seven mice were randomized into model group, Ruyanning group and Pingxiao group after inoculating MCF-7 cell. The conditions of drink and food, body weight and transplanted tumor weight were observed and recorded every other day , Spleen and tumor were taken and weighed when the mice were sacrificed in each group respectively on the eighth day. Then the inhibition rate of tumor and indices of spleen were calculated . All the tumor tissues were sliced and pathomorphological observed by transmission electron microscope (TEM).HER-2 protein expression of tumors tissues were detected by western blotting.Results:1. The decline of body weight in Ruyanning group were lower than the other two groups(p﹤0.01).2. Tumor weight of model group was significantly higher than the other two groups (p <0.01).3. The index of spleen in Ruyanning group was higher than Pingxiao group and the difference was significant(p﹤0.05).But comparing to the model group, there was no significant difference(p>0.05).4. The tumor cell apoptosis of the MCF-7 breast cancer could be induced by Ruyanning Decoction.5. The expreesion of HER-2 protein of the Ruyanning group and the Ping xiao group were both lower than the model group and the difference was signifi-cant (p﹤0.05).Conclusion: 1. The prescription of Ruyanning can improve body status of mice.2. The prescription of Ruyanning can enhance the index of spleen and improve immune function possibly.3. The prescription of Ruyanning can inhibit tuomor to some extent and the possible mechanisms of which are induced apoptosis and down- regulate expression of HER-2.
