黄芪及其有效部位干预化疗性贫血的实验研究

【摘要】 目的:本研究选用化疗药物环磷酰胺建立小鼠化疗性贫血的实验模型,观察黄芪及其有效部位对化疗性贫血模型小鼠血液毒性的干预作用,进一步探明黄芪各有效部位干预化疗血液毒性的差异性。材料与方法:根据外周血红、白细胞计数将132只SPF级BALB/c(雌雄各半)小鼠随机分为11组,即正常组、模型组、中药对照组、黄芪饮片组、糖苷酮组、糖苷组、糖酮组、酮苷组、多糖组、黄酮组、皂苷组,每组12只。正常组、模型组均按照0.2ml/20g体重剂量胃饲蒸馏水;中药对照组按照3ml/kg体重剂量胃饲黄芪精混悬液;黄芪饮片组按照9g/kg体重剂量胃饲黄芪水煎液;黄芪各有效部位组制备好的溶液按照9g/kg体重剂量(黄芪生药量)分别胃饲相应的治疗药物。每日一次,连续给药15天。于第1d、第3d、第5d,正常组除外,其余各组小鼠按照100mg/kg体重剂量腹腔注射环磷酰胺,总计3次,末次灌胃24小时后取材,检测小鼠外周血白细胞、红细胞、淋巴细胞、血小板计数、血红蛋白浓度以及胸腺和脾脏指数等各项指标。结果:1.模型组小鼠于造模第7d测外周血血红蛋白浓度,血红蛋白浓度下降正常的20%,表明化疗性贫血动物模型成功建立。2.各给药治疗组对环磷酰胺引起的小鼠精神萎靡,食少倦怠,形体消瘦,蜷缩弓背,行动迟缓,皮毛枯槁,稀松无华,对外界刺激反应迟钝,面、眼、唇、耳、尾呈苍白色等现象,均明显改善。3.与模型组比较,中药对照组、黄芪饮片组及其有效部位各组均能提高外周血白细胞计数;除黄芪饮片组外,其余各组均能提升模型小鼠外周血血红蛋白浓度;除黄芪饮片组、糖苷酮组和黄酮组外,其余各组还可增加模型小鼠外周血红细胞计数;除黄酮组外,其余各组均能提升模型小鼠外周血淋巴细胞计数;黄芪饮片组及其有效部位各组对血小板计数无影响。4.与模型组比较,中药对照组、黄芪饮片组、糖苷酮组、糖苷组、糖酮组、酮苷组、多糖组、黄酮组能增加模型小鼠的胸腺和脾脏指数,皂苷组仅能增加模型小鼠的脾脏指数。结论:1.黄芪及其有效部位对环磷酰胺所引起的血液毒性和免疫抑制等毒副作用有不同程度的拮抗作用。2.黄芪多糖和黄芪皂苷是促进红系造血的最佳有效部位;黄芪皂苷是促进白系造血的最佳有效部位。

【Abstract】 Purpose:To further study the variability of Astragalus mongholicus and its effective components in influencing hematotoxicity by observing the effectiveness on anaemic mice resulting from chemotherapeutics—CTX (cyclopho- sphamide).According to peripheral blood erythrocyte and leucocyte count, divided 132 SPF mice (half male) into 11 groups—normal control, model, TCM control, Astragalus mongholicus, glucoside and ketone group, glucoside group, saccharon, ketoside group, polysaccharide group, flavone group, saponin group, in randomication principle. Normal control group and model group was fed with 0.2ml/20g distilled water intragastrically; TCM control group was fed with 3ml/kg astragalus oral solution; Astragalus mongholicus group was fed with 9g/kg Astragalus mongholicus decoction, so do other astragalus effective components groups, for 15 days. At the 1th, 3th, 5th day, all groups except normal control one were intraperitoneally injected CTX with the dosage of 100mg/kg. 24 hours after the last intragastric administration, killed all the mice for getting materials which were used to test peripheral blood erythrocyte and leucocyte count, hemoglobin, lymphocyte, neutrophil, eosinophile granulocyte, blood platelets count, thymus and spleen index, and so on.Results:1. At the 7th d, peripheral hemoglobin concentration of the mice model group decreased to 20%, which indicated the BMD model was established.2. Symptoms of every treated groups resulting from CTX can relieve such as depression of spirit and decrease of activities, torpidity, decrease of weight, reducing of food and drink, the lack of shinning of fur, pale eye, lip, ere and tail, loosing stool.3. Comparing with model group, TCM contrel group, Astragalus mongholicus and its effective components can increase peripheral leukocyte count; every groups, but Astragalus mongholicus group, can increase hemoglobin concentration; except of Astragalus mongholicus, glucoside and ketone group, flavone group, can increase peripheral red blood cell count; except of flavone group, can increase peripheral blood lymphocyte count; Astragalus mongholicus and its effective components have no influence on blood platelets count.4. comparing with model mice,the thymus and spleen index was increased in TCM control,Astragalus mongholicus, glucoside and ketone group, glucoside, saccharon, ketoside group, polysaccharide group, flavone group. saponin group can only increase the spleen index.Conclusion:1.Astragalus mongholicus and its effective components have the antagonistic action for hematotoxicity and immunity depressed resulting from CTX in different degree.2. Astragalus polysaccharide and astragalus are the most effective part of improving erythropoiesis; Astragalus astragalus is the most effective part of improving leukon.
