太白米的生药学和化学成分研究

【摘要】 太白米Notholirion bulbuliferum (Lingelish.) Stearn.又名假百合,为百合科(Liliaceae)假百合属(Notholirion)植物假百合的干燥小鳞茎。主要分布于我国陕西、西藏、四川、云南等省,是具有悠久药用历史的名贵中草药。太白米性温、味辛、微甘苦,具有宽胸利气、健胃、镇痛等功效,主治气滞心痛、胃腹胀痛、呕吐、风寒咳嗽等,此外还具有抗肿瘤、镇痛等药理作用[1-7]。本论文对太白米的生药学特性和化学成分组成进行了系统研究,并对有机酸类化合物的研究情况作了简要综述。本课题采用肉眼、显微观察、照相与绘图相结合的方法对太白米原植物形态、药材性状、组织及粉末特征进行了全面研究,确定了其生药学特性,为其性状和显微鉴定提供了依据;并对太白米的水分、总灰分、酸不溶性灰分及浸出物进行了初步研究。同时采用系统溶剂提取法、薄层色谱法、硅胶柱层析法、LH-20柱分离法、C18反相柱分离法等对太白米的化学成分进行了系统研究,从太白米中分离得到了1、2、3、4、5,五个单体化合物,并运用化学定性鉴定和光谱测定(MS、1H—NMR、13C—NMR)确定化合物1为β-谷甾醇(β-sitosterol);化合物4为胡萝卜苷(daucosterol);化合物2、3、5均为有机酸类成分,分别为:对甲氧基桂皮酸(2-Propenoic acid,3-(4-methoxyphenyl))、对羟基桂皮酸(2-Propenoic acid,3-(4-hydroxyphenyl))、对羟基桂皮酸酯苷(β-D-Glucopyranose,1-[3-(4-hydroxyphenyl)-2-propenoate] ),其中2,3,5为首次从植物中分离得到的天然产物,丰富了天然产物的种类,具有重要意义。

【Abstract】 Taibaimi aslo name as Notholirion,derived from the little bulb of Notholirionbulbuliferum (Lingelish.) Stearn. ,is one of the valuable Traditional Chinease Medicines (TCM) that has a long medical history. Taibaimi mainly grown in theprovinces of Shannxi,Xizhang,Sichuan and Yunnan. Taibaimi is walm in nature and little bitter in taste;has effect of relaxing the chest,invigorating stomach and analgesic. For treatment of stagnation of vital energy and epigastragia; distension and ache of stomach and abdominal;vomit;cough of wind-cold.Moreover,Taibaimi has pharmacolo-gical effect of antitumor,pain-alleviating[1-7].The thesis has made comprehensive pharmacognosy characteristics and chemical composition study of Notholirion hyacinthinum ( wils.)stapf .which come from Lilium Liliaceae Notholirion, and make a review of organic acid compounds.The issue through the methods of eye and microscopic observation,p-hotography and drawing combining,we have determined it’s pharmacogon-osy by making a comprehedsive study of original plant morpgology of Taibaimi and it’s organizations and powder characteristics,medicinal traits.Through this study we provided basis for its properties and micoscopic identification.Acorrding to the Phramacopoeia methods of water、ash and extact determination,we have set the content standards of Taibaimi’s water, total ash, acid insoluble ash and extacts.Using the system solvent extraction, TLC, silica gel column chromatography, LH-20 column separation to make systematically study of chemical constituents of Taibaimi,isolated five pure compounds(1,2,3,4,5) from Taibaimi.We cocluded the five compounds structure by using chemical identification and spectrometric determination (MS, 1H-NMR, 13C-NMR),identified compound 1 asβ-sitosterol(β-sitosterol);Compound 2 daucosterol(daucosterol);compound 3,4,5 are the organic acids, respectely:p-methoxy cinnamic acid (2-Propenoic acid,3-(4-methoxyphenyl)),hydroxy cinnamic acid (2-Propenoic acid,3-(4-hydroxyphenyl)),hydroxy cinnamid acid ester glucoside (β-D-Glucopyranose, 1-[3-(4-hydroxyphe-nyl)-2-propenoatel.
