电针对脑缺血再灌注模型大鼠总抗氧化能力、cAMP、cGMP调节机理的研究

【摘要】 观察电针对局灶性脑缺血再灌注大鼠海马、肝脏T-AOC、cAMP、cGMP的影响,探讨脑缺血再灌注损伤的发病机制以及电针对脑缺血再灌注损伤的抗氧化保护机理,为进一步推广电针在缺血性脑中风防治中的临床应用提供科学证据。采用雄性SD大鼠运用大脑中动脉栓塞法建立大鼠脑缺血再灌注模型,随机分为模型组、电针治疗组和假手术组。采用Julio氏神经行为学评分法评价神经功能缺损状况,通过比色法观察各组变化,用放射免疫法分析各组缺血区海马及肝组织cAMP、cGMP的变化,并对cAMP/cGMP比值的变化进行比较分析。与假手术组比较,其余各组评分均明显降低;电针组与模型组比较,评分有明显性差异(P<0.01)。脑缺血再灌注后缺血区海马及肝组织中T-AOC含量减少,电针治疗组与脑缺血组相比,T-AOC含量上升。脑缺血再灌注后,缺血区海马及肝组织中cAMP含量降低,cGMP含量升高,cAMP/cGMP比值减小,经电针治疗后的电针组cAMP含量显著上升,cGMP含量显著下降,cAMP/cGMP比值增大,与模型组比较,差异均有统计学意义。1.电针能够在一定程度上改善脑缺血再灌注损伤,使大鼠神经功能缺损得到相应的纠正。2.电针对大鼠脑缺血再灌注损伤的抗氧应激保护效应,可能是通过提高机体抗氧化的能力,调节细胞信号转导中第二信使cAMP、cGMP,抑制缺血再灌注肝脏及缺血神经元的进一步损伤,从而发挥机体的康复保护作用。

【Abstract】 Stroke is one of the major diseases that damage severely human health. With the advent of aging society, the incidence,mortality and disability rates of stroke are gradually the highest among all the diseases,and acupuncture treatment of stroke has the exact effect. In order to discusses the effect of electro-acupuncture on acute focal cerebral ischemia-reperfusion injury in rats,and research the effect of EA on T-AOC、cAMP and cGMP.we use the rat model of focal cerebral ischemia-reperfusion in the experiment,observe the level of T-AOC、cAMP and cGMP,The results are:1.Electro-acupuncture can improve cerebral ischemia-reperfusion injury in rat,and the defect on neuropathology can be corrected on some extent.2.The mechanisms of electro-acupuncture on rat cerebral ischemia-reperfusion injury was possibility that the acupuncture can enhance the ability of oxidation resistance,and adjust the second messenger—cAMP,cGMP in cell signal transduction.The balance between cAMP and cGMP to be restored,in order to play a therapeutic effect.
