狼毒(月腺大戟)炮制机理研究

【摘要】 狼毒(月腺大戟)炮制原理研究为国家中医药管理局有毒中药炮制技术与原理研究课题,本课题主要通过对狼毒化学、药理等方面的研究,来探讨狼毒的炮制原理,为制定狼毒炮制品的质量标准提供理论依据。主要研究内容:一、文献研究对历代医药文献及全国各省市中药炮制规范中有关狼毒炮制资料进行了整理分析,总结了狼毒炮制的历史沿革,为其炮制工艺改进提供了思路和方法,为研究狼毒的炮制原理奠定理论基础。二、化学研究采用HPLC法对炮制前后狼毒提取液进行检测。通过图谱比较研究,指出了生品与炮制品在HPLC色谱图上的不同。对狼毒炮制前后狼毒乙素、狼毒丙素、多糖的含量进行了测定,应用GC-MS对狼毒炮制前后脂肪油中的化学成分进行了分析比较。对狼毒生品的化学成分进行了系统的分离,从狼毒的石油醚提取部分和乙酸乙酯提取部分分离得到十六个化合物,分别是:狼毒乙素、狼毒丙素、阿魏酸二十八酯、没食子酸、β-香树脂醇乙酸脂,β-谷甾醇,2,5-呋喃二乙酸、阿魏酸、对羟基苯甲酸、豆甾醇、胡萝卜苷、棕榈酸、硬脂酸、1,10-二十九二醇及两个新化合物:2,4-二羟基-6-甲氧基亚甲基-3-甲基苯乙酮和该化合物的苷。三、药理研究通过对比狼毒生制品急性毒性、测定对二甲苯致小鼠耳廓肿胀影响,采用体外方法评价其对DPPH自由基的清除能力及采用MTT法进行体外抗肿瘤作用的比较,为阐明狼毒的炮制原理提供了实验依据。

【Abstract】 The processing principle research for Euphorbia ebiacteolata Hayata.was one of the topics named as "study of Processing Techniques and Principles of toxic drugs" of the State Admini-stration of Traditional Chinese Medicine, which discussed the processing principle of langdu, mainly through the aspects of chemistry, pharmacology, etc., to provide the theoretical basis for making the quality standard of the processed sample. Main research contents:1.Literature researchThrough the study on medical literatures about Euphorbia processing from past dynasties and the standards of herbal processing practiced in different districts in China, the author summed the history of Euphorbia processing, and its original meaning and gave suggestions to improve the Euphorbia processing and lay a theoretical foundation for the processing principle of langdu.2.Chemical researchWe assay the extraction liquid for the raw and processed samples of langdu with HPLC un-der different absorption wavelengths.The result of the contrasted altas analysis manifested that changes occurring to the content.The content of 2,4-dihydroxy-6-methoxy-3-methyl-acetophenone; 2-hydroxy-6-methoxy-3-methyl-acetophenone-4-β-glueoside and Polysaccharide were deter-mined before and after langdu was processing. Making use of the GC-MS to compare the chemical composition in the expressed oil before and after processed of euphorbia.The chemical compounds of the processed langdu were systemically separated, 2,4-dihydroxy-6-methoxyl-methylene-3-methyl-acetophenone,2,4-dihydroxy-6-methoxyl-methyl ene-3-methyl-acetophenone-4-O-β-D-glucopyranoside,octacosyllate,3-acetyl-β-amyrin,3,3’-diac etyl-4,4’-dimethoxy-2,2’,6,6’-tetrahydroxy diphenylmethane,2,4-dihydroxy-6-methoxy-3-methyl acetophenone, gallic acid,2-hydroxy-6-methoxy-3-methyl-aeetophenone-4-β-glueoside, P-sitosterol and daucosterol;ferulic acid,4-hydroxybenzoic acid,β-stigmasterol, hexadecanoic acid were respectively received from the petroleum and ethyl acetate extract of langdu.3.Pharmacological researchA method was established to compare activities of acute toxicity, anti-inflammatory, anti-oxidant and MTT method was developed to compare the activity of antitumor in vitro of dief-frent raw and processed samples of langdu,which also provided the scientific basis for clarifying the processing principle of langdu.
