规模化蛋鸡舍消毒剂的筛选及空气消毒规程的研究

【摘要】 目的:通过测定15种消毒剂(百毒杀、新大卫、PVP碘、过氧化氢、84、百疫杀、百毒杀、,百毒强杀、万仑菌清、新洁尔灭、复合酚、碘附、菌毒灭、弱酸性离子水、电解次氯酸钠)的抑菌能力和杀菌能力,筛选出抑菌能力和杀菌能力强的消毒剂,随后使用筛选出的消毒剂建立夏季和冬季规模化蛋鸡舍的消毒规程,并且对消毒效果的影响因素设计正交分析,选择出最佳的组合,制定最适合规模化蛋鸡舍的消毒规程。方法:采用肉汤稀释法测定15种消毒剂对标准菌株(大肠杆菌8099,金黄色葡萄球菌ATCC6538,枯草杆菌黑色变种芽孢ATCC9372)和鸡舍中分离的菌株(19株大肠杆菌,16株金黄色葡萄球菌与7株枯草芽孢杆菌)的最小抑制浓度(MIC)和最低杀菌浓度(MBC);悬液定量杀菌试验测定15种消毒剂对3种标准菌株的杀灭效果;采用自然沉降法测定规模化鸡舍环境细菌总数;正交试验分析研究各因素对消毒效果的影响。结果:通过对15种消毒剂MIC值、MBC值和杀灭对数值的综合分析,从中筛选出杀菌能力和抑菌能力均强的消毒剂有:百毒杀S、弱酸性离子水、电解次氯酸钠、新大卫、新洁尔灭、百疫杀、碘附。夏季的消毒程序建议:未产蛋鸡舍使用312.5mg/L百毒杀S,喷雾50ml/m3,间隔108h消毒一次;产蛋鸡舍使用625mg/L的百毒杀S,喷雾30ml/m3,间隔84h消毒一次。冬季的消毒程序建议:产蛋鸡舍625mg/L的百毒杀S,喷雾18ml/m3,间隔66h消毒一次。通过对影响消毒效果的因素进行极差分析和方差分析,夏季的试验设计中的消毒剂种类和消毒剂浓度对消毒剂效果的影响极显著(P<0.01),喷雾量对消毒效果的影响不显著(P>0.05),最佳的消毒程序组合为:百毒杀S稀释至625mg/L,喷雾为50ml/m3,消毒间隔时间90h。冬季的试验设计中喷雾量对消毒效果的影响显著(P<0.05),消毒剂种类和消毒剂浓度对消毒效果的影响极显著(P<0.01),消毒时间对消毒效果影响不显著(P>0.05),最佳的消毒程序组合为:百毒杀S稀释至625mg/L,喷雾为24ml/m3,消毒时间为30min,消毒间隔时间72h。

【Abstract】 The aim:By testing bacteriostasis and disinfection capability of 15 kinds of disinfector-Didecyl dimenthl ammonium bromide solution10%, Dilute glutaral solution, 5% Povidohe Iodine, H2O2,84, Hydrocarbon dinethy ammonium chloride compound glutaral soluation, Benzalkonium bromide solution,20g/L Povidone Iodine solution, For-maldehyde solution, Bromogeramine,45% complex phenol,3% Povidone Iodine,41 %complex phenol, Weak acid hypochlorous water, Electrolyzed oxidizing water-we could screen out disinfector of strong bacteriostasis and disinfection capability. Then we set up Codes for disinfenction of large scale laying house in summer and winter by using screened disinfector, design perpendicular analysis for factor of disinfective effectivenes-s, choose optimal combination and institute effective regulation which is most suited for large scale laying house. The method:Using broth dilution method to test the minimum inhibitory concentration(MIC) and minimum bactericidal concentration(MBC) of 15 kinds of disinfector toward standard strain and strain from laying house, using suspension quantitation bactericidal trial to test disinfective effectiveness of 15 kinds of infector toward 3 kinds of standard strain, using natural sedimentation to test total viable organism of large scale of laying house and using orthogonal experiment to analyze and research the effectiveness of each factor toward disinfective effectiveness.The result:Using analysis by synthesis of 15 kinds of disinfector MIC, MBC and disinfective effectiveness, we could screen out disinfector of strong disinfection and bacteriostasis capability, such as Didecyl dimenthl ammonium bromide solution10%, Weak acid hypochlorous water, Electrolyzed oxidizing water, Dilute glutaral solution, Bromogeramine, Hydrocarbon dinethy ammonium chloride compound glutaral soluatio -n,3% Povidone Iodine. Advice for disinfector program in summer:Using 312.5mg/L Didecyl dimenthl ammonium bromide solution 10% in non-laying house to spray 50ml/m3 and disinfect once every 108h. Using 625mg/L Didecyl dimenthl ammonium bromide solution10% in laying house to spray 30ml/m3 and disinfect once every 84h. Advice for disinfector program in winter:Using 625mg/L Didecyl dimenthl ammonium bromide solution 10% in laying house to spray 18ml/m3 and disinfect once every 66h. Using analysis of range and analysis of variance for factor of disinfective effectiveness, we could find that the influence of the kinds and concentration of disinfector toward disinfective effectiveness in summer design of experimentis great significant (P<0.01). But the influnce of the spraying amount toward disinfective effectiveness is not significant (P>0.05). The optimal combination of disinfective program is that we should dilut Didecyl dimenthl ammonium bromide solution 10% to 625mg/L, spray 50ml/m3 and disinfect once every 90h. Furthermore, we could find that the influence of spraying amount toward disinfective effectiveness in winter design of experiment is significant (P<0.05), the influnce of the concentration and the kinds of disinfector toward disinfective effectiveness is much significant (P<0.01) and the influnce of disinfective time toward disinfective effectiveness is not significant (P>0.05).The optimal combination of disinfective program is that we should dilut Didecyl dimenthl ammonium bromide solution 10% to 625mg/L, spray 24ml/m3, disinfect by 30 min and disinfect every 72h.
