三种中国南海红树林内生真菌次级代谢产物的研究

【摘要】 近年来,海洋微生物次级代谢产物成为国内外科学家们研究的热点。海洋微生物生存环境复杂、代谢方式独特,能产生结构新颖、具有药理活性的次级代谢产物。其中红树林真菌由于资源丰富,产生活性代谢产物的能力强,是目前研究得最系统和最全面的海洋真菌。本论文运用多种色谱技术及现代波谱技术,系统研究了来自中国南海红树林的三种内生真菌SK3RW6M、ZZF42、SKX1YW3G1的次级代谢产物。通过薄层层析、柱层析、凝胶柱色谱、高效液相色谱等分离方法及MS、1D/2D NMR等波谱技术,共分离鉴定15个化合物,包括2个未见报道的新xanthone类化合物6M-1和6M-2。其中,从真菌SK3RW6M的菌液及菌体中分离得到5个化合物,分别为6M-1(新化合物)、6M-2(新化合物)、麦角甾醇、过氧麦角甾醇、甘露醇;从真菌ZZF42的菌液及菌体中分离得到5个化合物,分别为xylopyridine A、14-甲基十五碳酸甲酯、硬脂酸、硬脂酸甲酯、正二十四烷酸;从真菌SKX1YW3G1的菌液及菌体中分离得到5个化合物,分别为5-丁基-2-吡啶甲酸(镰刀菌酸)、5-丁基-2-吡啶甲酸甲酯(镰刀菌酸甲酯)、环(苯丙-苯丙)二肽、环(甘-苯丙)二肽、胸腺嘧啶。论文还对近年来从天然生物资源中分离得到的xanthone类化合物进行综述,阐述了新化合物的结构和相关药理活性。共引用化合物147个,有生物活性报道的化合物85个;这些新化合物中,来源于海洋微生物的有14个,其中12个生理活性显著。

【Abstract】 In recent years, researches on secondary metabolites of marine microorganisms have become hotspots that domestic and oversea scientists pay much attention to. Marine microorganisms could produce numerous novel and bioactive compounds because of the complicated living environment and their peculiar metabolism. Mangrove-derived fungi are rich in resources and had a great capacity for producing bioactive metabolites, which make them the most systemically and roundly researched marine fungi in present.On the basis of several chromatography techniques and spectral methods, 3 species mangrove fungi SK3RW6M, ZZF42 and SKX1YW3G1 collected from South China Sea were studied on their secondary metabolites in this thesis. 15 Compounds including 2 new compounds with xanthone structure were isolated using thin-layer chromatography, column chromatography, Sephadex LH-20 and HPLC, characterized by MS, 1D NMR and 2D NMR. Compounds isolated from the culture medium and mycelium of fungus SK3RW6M are 6M-1(new), 6M-2(new), ergosterol, ergosterol peroxide and Mannitol. Xylopyridine A,methyl 14-methylpentadecanoate, stearic acid, methyl stearate and tetracosanoic acid were isolated from the culture medium and mycelium of fungus ZZF42. There are also 5 compounds isolated from the culture medium and mycelium of fungus SKX1YW3G1, including 5-butylpicolinic acid, methyl 5-butylpicolinate, 5-methylpyrimidine-2,4(1H,3H)-dione, 3,6-dibenzylpiperazine-2,5-dione and 3-benzylpiperazine-2,5-dione.Besides that, the thesis also reviewed new compounds of xanthone derivatives isolated from natural organism resources. 147 compounds are cited, among which 84 compounds were reported with biological activities. 14 new compounds come from marine-derived fungi, of which 12 have excellent pharmacology activities.
