三种喹诺酮类抗生素对斜生栅藻的毒性效应

【摘要】 盐酸恩诺沙星(ENFX)、盐酸环丙沙星(CPFX)和氧氟沙星(OFLX)是常用于水产养殖的抗生素药物。它们在作用于靶生物的同时,也会对水生生态系统中的其它非靶生物的生长和生理产生影响。本研究以斜生栅藻(Scenedesmus obliquus)为实验生物,研究不同浓度的ENFX(0,24、48、80、96、120mg/L)、CPFX(0、14.6、29.2、43.8、73、109.5mg/L)和OFLX(0、22、44、66、112、168mg/L)对斜生栅藻的毒性效应。研究结果表明:ENFX、CPFX和OFLX对斜生栅藻均属于低毒物质。它们的96h EC50分别为59.44mg.L-1、51.71mg.L-1和134.07mg. L-1,安全浓度分别为5.94mg.L-1,5.17 mg.L-1和13.41 mg.L-1。ENFX、CPFX和OFLX暴露下,光合色素含量均降低,其中叶绿素a的含量远高于叶绿素b和类胡萝卜素。ENFX暴露下,可溶性蛋白含量先升高后降低,而CPFX和OFLX暴露下,可溶性蛋白均降低,且CPFX处理降低的幅度更大。ENFX、CPFX和OFLX暴露下,超氧阴离子(O2-)的产生速率均随着处理浓度的增加而加快,且高浓度组产生速率远高于低浓度组。低浓度组处理下,MDA含量的变化均不大,高浓度时,含量则明显高于对照组。ENFX和CPFX暴露下,SOD活性随着处理浓度的增加先激活后抑制,而OFLX暴露下,则一直处于激活状态。ENFX、CPFX和OFLX暴露下,脯氨酸变化规律相似。低浓度时,脯氨酸含量与对照相比变化不大,甚至还有所降低,而高浓度处理,则上升迅速,出现大量积累。

【Abstract】 Hydrochloric acid enrofloxacin (ENFX), ciprofloxacin hydrochloride (CPFX) and ofloxacin (OFLX) are antibiotics which commonly used in aquaculture. They not only act on target organisms, but also on the other non-target organisms in quatic ecosystems,and affect their growth and physiology. In this study, Scenedesmus obliquus is experimental organism. The toxic effects of different concentrations of ENFX (0,24,48,80,96,120 mg/L),CPFX(0,14.6,29.2,43.8,73,109.5 mg/L) and OFLX (0,22,44,66,112,168 mg/L) on Scenedesmus obliquus were studied in present study.The results showed that:To Scenedesmus obliquus , all of the three antibiotics were low-toxic.The 96h EC50 of ENFX, CPFX, and OFLX were 59.44mg.L-1, 51.71mg.L-1, and 134.07mg.L-1 respectively.The security concentrations of them were 5.94mg.L-1, 5.17 mg. L-1 and 13.41mg.L-1,respectively.Exposured to ENFX, CPFX and OFLX ,the photosynthetic pigment content of Scenedesmus obliquus decreased, and the content of chlorophyll a were much higher than that of chlorophyll b and carotenoids.Treated with ENFX , the soluble protein content of Scenedesmus obliquus increased and then decreased, whereas with CPFX and OFLX, decreased, and the range of CPFX treatment to reduce more.Exposured to ENFX, CPFX and OFLX,the superoxide anion (O2-) production rates increased with the concentration accelerating the production rate and high concentration much higher than the lower concentrations. Treated with lower concentrations of the three quinolone antibiotics , the contents of MDA had a minor change , while with high concentration, changed significantly. Exposured to ENFX and CPFX, the activity of SOD increased with the concentration of treatment, while to OFLX, remained active in all concentrations.Exposured to ENFX, CPFX, and OFLX,the contents of proline similar variation. Compared with the control group,the contents of proline changed little in the low concentration group, or even reduced, while in the high concentration group, then increased rapidly, a large number of accumulation.
