异噁唑-5-酮及吡唑-5-酮衍生物的一锅法及绿色合成

【摘要】 有机杂环化合物是近年来有机化学领域发展较快的一部分,不但在理论上,而且在实际应用上都有很重要的意义。异噁唑、吡唑、噻唑等含氮杂环化合物是一类具有广泛生理活性的化合物,在医药和农药方面有着广泛的应用,是当今药物研究的一大热点。查尔酮及其衍生物是一种重要的有机中间体,广泛地用于有机合成和药物合成当中。因查尔酮具有多个反应活性中心,在配位化学和超分子化学中具有广泛的应用。这类杂环化合物的生物活性主要表现在抗菌、消炎、止痛、抗焦虑、抗癌等,还可作为杀菌剂、除草剂、杀虫剂、植物生长调节剂等,因此,受到了许多药物化学家和农药学家的关注。为寻找新合成路线和新方法,我们用一锅反应和绿色合成等合成方法,合成了未见文献报道的取代异噁唑-5-酮,吡唑-5-酮衍生物。本论文分为以下两个部分:第一部分为文献综述部分,对异噁唑、吡唑、噻唑及其衍生物的生理活性、重要合成法和化学性质在国内外的研究状况,作了简要的阐述。第二部分为实验部分,主要工作分为四个部分:1. 4-芳亚甲基-4,5-二氢-3-苯基异噁唑-5-酮的一锅法及绿色合成2. 4-芳亚甲基-4,5-二氢-1,3-二苯基吡唑-5-酮的一锅法及绿色合成3. 4-取代-4,5-二氢-3-异丙基异噁唑-5-酮和4-取代-4,5-二氢-3-异丙基-1-苯基吡唑-5-酮的一锅法合成4. N-(1,3-二苯基-5-氯-吡唑-4-基)-N′-(芳基噻唑-2-基)-甲醛腙和N-(4-芳基噻唑-2-基)-对苄氧基苯乙酮腙的合成

【Abstract】 In recent years, organic heterocyclic compounds have been developed rapidly in the field of organic chemistry, and take an important signification not only in theory but also in practical application aspects. The heterocyclic compounds containing nitrogen such as isoxazole, pyrazole, thiazole are some important compounds of biological ac tivity and play an extensive application in modern medicine and pesticide aspects, is a focus of the research of drug nowadays. Chalcone and its derivatives are important organic intermediates, and widely used in coordination chemistry and super-molecular chemistry. As a kind of versatile intermediates, chalcones have been used widely in the synthesis of organic compounds and medicines.There are mostly biological activities, such as antibacterial, antiphlogistic, acesodyne, anticonvulsant, anticancer, bactericide, herbicide, pesticide and plant growth regulator and so on, so many drug chemists and pesticide learners pay attention to these heterocyclic compounds. In view of new synthetic method, the novel derivatives of isoxazole-5-one, pyrazole-5-one were synthesized by one-pot synthetic method and green procedure.The thesis includes two major parts:The first part is about literature review. The important biological activity, synthetic methods and chemical characteristics of the isoxazole, pyrazole and thiazole derivatives that have been studied in china and other countries were explicated briefly.The second part is about experimental content. Major work as follows:1. One-pot synthesis of 4-arylmethylidene-4,5-dihydro-3-phenyl isoxazol-5-ones2. One-pot synthesis of 4-arylmethylidene-4,5-dihydro-1,3-diphenyl pyrazol-5-ones3. One-pot synthesis of novel 4-arylmethylidene-4,5-dihydro- 3-isopropylisoxazol– 5-ones and 4-arylmethylidene-4,5-dihydro-1-phenyl-3-Isopropylpyrazol-5-ones4. Synthesis of N-(4-1,3-diphenyl-5-chlor -pyrazol-5)-N’-( phenyl-thiazole-2-) N-(1,3-diphenyl-5-chloro-pyrazole-4-yl)-N’-(arylthiazol-2-yl)-formylhydrazone and N-(4-arylthiazol-2-yl) -benzyloxy acetophenone hydrazone
